Background and Objective: Granulosa cell tumors of the ovary are rare neoplasms that originate from sex-cord stromal cells. They are characterized the endocrine activity and represent 2-3% of malignant tumors of the ovary.The molecular pathogenesis of granulosa cell tumors is not known.In the female gonad, FOXL2 gene is essential for granulosa cell differentiation and ovary maintenance.The P450arom catalyzes three consecutive hydroxylation reactions and converts C19 androgens to estrogen. It is a key enzyme for estradiol biosynthesis from ovarian granulosa cells. Ki67 is a cell proliferation nuclear antigen,which is recognized as the most ideal marker reflecting cell proliferation level. The aberrated ratio of cell proliferation and apoptosis is a vital mechanism that leads to the development of cancer.The purpose of the study is to define the role of Ki67,FOXL2 and P450arom during the evolution of ovarian granulosa cell tumors.Method: The expression of proliferation cell nuclear antigen Ki67, FOXL2 and P450arom was measured in 37 cases of ovarion granulosa cell tumors tissues by immunohistochemical method. The relationship between the expression of FOXL2 , P450arom ,Ki67 and clinical pathological parameters that of the expression of these protein were analysed.Result: The expression of Ki67 , FOXL2 and P450arom in ovarion GCT were higher than that in the normal tissues. The expression of Ki67 gradually increased in clinical stageⅠ,Ⅱ,Ⅲ. The expression of FOXL2 in clinical stageⅡ+Ⅲwas higher than in clinical stageⅠ.The expression of Ki67 in the of karyokinesis≥3/10HPF was higher than that in the karyokinesis <3/10HPF. The expression of FOXL2 in the granulosa cell tumors of diffuse(solid)and other pattern was higher significantly than in follicular pattern.A significantly positive relationship was observed between expression of Foxl2 ,P450arom and Ki67 protein in GCT.Conclusion: FOXL2 and Ki67 can be served as subsidiary markers of the tumors,and their expression can provide a theoretical basis for predicting biological behavior of ovarion GCT.
