Aim: To investigate the anti-tumor effect of beta-aescin and its mechanism.Methods:①MTT assay was used to select sensitive cells line in SGC-7901,SMMC-7721,HL-60,M-231,H1299; MTT assay, 3H-TdR and colony-forming units assay were used to determine inhibitory effects of beta-aescin in vitro.②Anti-migration effects of beta-aescin were observed by scratch assay and adhesion assay.③Change in form of cells treated with beta-aescin was observed by Giemsa staining; Fas expression, cell cycle and apoptotic rate were analyzed by FCM; Western blot was employed to analyze the expression of bcl-2 and bax protein.④Immunosuppressive model in mice was established via intraperitoneal injection of cyclophosphamide and irradiation of 60Co-γray. The effects of beta-aescin were evaluated by immune organ indexes, phagocytic percentage of macrophage, hemolysis of cock red blood cells and the proliferation of spleen lymphocytes.Results:①Results showed that beta-aescin could obviously inhibit the proliferation of SGC-7901 and SMMC-7721 cells in vitro.②Results showed that beta-aescin had an anti-metastasis effect in vitro.③Apoptotic features of cells cultured with beta-aescin could be found by Giemsa staining. Beta-aescin could induce SGC-7901 cell apoptosis and up-regulate the Fas expression. SGC-7901 cells were inhibited in G0/G1 and S phase. Western blot suggested that beta-aescin could up- regulate the bax expression and down-regulate the bcl-2 expression.④Beta-aescin could increase the thymus and spleen indexes, phagocytic percentage of macrophage, quantitative hemolysis of cock red blood cells and the proliferation of splenic lymphocyte transformation rate.Conclusion: The results suggest that beta-aescin has significant inhibitive effect on SGC-7901. The mechanisms might be due to the inducing apoptosis of SGC-7901, anti-metastasis and improving immunomodulatory effects of beta-aescin.
