1.Objective:To observe the change of NO,NOS,L-selectin and connexin43(Cx43) expression in rat renal ischemic reperfusion injury after renal ischemic preconditioning or Danshen preconditioning and investigate the effect of ischemic preconditioning on renal ischemic reperfusion injury.2.Methods:40 SD rats were randomly divided into 4 groups:sham-operated group(Sham-group),ischemic-reperfusion group(IR -group),ischemic preconditioning group(IPC-group),Danshen group(DS-group).The renal underwent 60-minute ischemic and 2-hour reperfusion.The level of Superoxide Dismutase(SOD),Malondialdehyde(MDA),NO,NOS of kidney,blood in each group were examined.The number of PMNs and Paller's count to assess the changes in renal pathological morphology. L-selectin expression in kidney was tested with immunohistochemichemistry.The expression levels of Cx43 protein of renal was examined by western blot analysis..3.Results:TNOS,INOS,NO,SOD:The values in IR-group were significantly lowerer than that in Sham-group,The values in IPC-group,DS-group were significantly higher than that in IR- group;MDA:The values in IR-group were significantly higher than that in Sham-group,The values in IPC-group,DS-group were significantly lower than that in IR-group; The numbers of PMNs and Paller's count:The values in IR-group were significantly higher than that in Sham-group,The values in IPC- group, DS-group were significantly lower than that in IR- group;The expression of L-selectin:The values in IR-group were significantly higher than that in Sham-group,The values in IPC- group,DS-group were significantly lower than that in IR-group;Protein expression of Cx43 in IR-group was significantly decreased as compared with that of Sham-group;The protein expression in IPC- group,DS-group were was significantly increased as compared with that of IR-group.4.Conclusion:Ischemic preconditioning and Danshen preconditioning could protect kidney from ischemic reperfusion injury,the mechanism might be that increase the protein expression of Cx43 in renal tissue and increase NOS in kidneys and blood,so that the expression of L-selection was inhibited.These improve the SOD's activity,reduce the levels of MDA in kidney and blood.
