Pathological change caused by large area burn not only can take place on constitution and organ impaired by heating power directly,such as cutaneous and subcutaneous etc.but also can cause a series of change in each organ all over the body.MODS have been the main cause of death in the fire burn patients.At present, we have not gotten the public special therapeutic method about the MODS,the main therapia still was prevention.Lately,many research indicated that,earlier period enteral nutrition had visible protection on the mucous membrane of the gastrointestinal tract after burning,which can prevent stress ulcer,gastrointestinal bleeding and enteric becteria shifting,can depress the atabolism caused by burn obviously,improve the nutritional status all over the body.The major immunutrition praeparatum researched by scholar was glutamine and arginine etc.there were materials confirmed that Gly can oppose endotoxemia,hemorrhagic shock,toxic shock and sepsis etc.lesion,can improve antiinflammatory /inflammatory immune disequilibrium caused by LPS etc.can against MODS generated possibly.Objective:explore the effection of glycine in opposing endotoxemia and protecting organic function in burn rats.Methods:1.The experimental animal of making model and group.Toally 96 wistar rats of either gender:the depilatory area was immersed into boiled water bath for 20% TBSA superficial second degree burn injury.Recover all rats by sodium lactate ringer's injection injected into the abdominal cavity of the rats(40ml/kg)6h later. Injected LPS 10mg/kg 3h later,and gave 1mg/3ml GLN or Gly to difference groups.The rats were sacrificed at 3,6,12,24h.All 96rats were randomly and equally divided into 4 groups:simple burn group(A),sepsis group(B),Gln Group(C),Gly Group(D),And each group was regrouped for 3,6,12,24h(n=6).2.Determination of endotoxin level of the experimental animal's blood plasm:The chromogenic substrate limulus amebocyte lysate(LAL) apply to detect the level of LPS in serum of each group rats.3.Determination of the experimental animal's liver and renal function:the animal's blood serum was determinated the level of BUN,Cr,ALP,ALT Bby automatic biochemistry analyzer.4.Detection of the pathomorphology of the organize:recipe the lung,liver and renal of the animal that was killed livingly,fixed by 15%formalin,then fixed four hours by AF,gradually dehydrated by 80%alcohol four hours,95%alcohol ten hours,dehydrated alcoholⅠ,dehydrated alcoholⅡand acetone each one hour; transparent by xyleneⅠand xyleneⅡeach forty minutes;steeped cere and embedded.Eventually,the paraffin block were incised into 5urn thickness section by Leitzll 516 slicing machine,routine HE dyeing,then observe the appearance change of the organize by light microscope.Results:1.Changes of LPS in different group:LPS level in every group was begin raising from 3h,and declined at 24h.Group A reached peak at 3h,group B at 6h,group C,D both at 12h,and 24h was lower than 6h in group B(P<0.05).The level of LPS in group B was dramatically higher than each other group at each time point(P<0.05),and group C was not significantly different from group D.2.Changes of organ function in different experiment condition:After burning,the liver and renal function of the rats was little abnormal during the first 24 hours. And the liver and renal function was significantly abnormal after the rats were assaulted by LPS,however,the function was improve after intra-gastric administration with Gin or Gly when the rats were assaulted by LPS.3.Changes of organ's histomorphology in different experiment condition:The general pathomorphology change of each organ showed interstitial substance inflammatory reaction after burned 24 hours.The interstitial substance vessel of lung dilat,part alveolar septum broaden,among which neutrophil infiltrate; hepatic congestion,part hepatic cell vescuolar degeneration,sinus hepaticus dilat,congestion,central vein congestion;renal interstitium vessal broaden, congestion,part renal cells degeneration,individual acinus renis analosis,the pathomorphology of each organ changed obviously aggravate after assaulted by LPS,and the change was significantly lessen after intra-gastric administration with Gln or Gly when the rats were assaulted by LPS.There are not obvious difference between Gln and Gly.Conclusions:1.The animal model of endotoxin after 20%TBSA superficial second degree burn injury was made successfully.The level of LPS,BUN,Cr,ALT,ALP of serum in the experimental group(B,C,D group) was obviously higher than the control group(A group) at each time point(P<0.05).2.Gln,Gly can protect the organs.They can depress the LPS level of the sepsis rats' blood serum and organs,and protect each organ's fuction.
