Study The Effect Of CD34~+CD38~-Leukemic Stem Cells And Nuclear Factor Kappa B By Traditional Medicine Fomular

Objective:To approach the effect of Leukemic stem cell (LSC) CD34~+CD38~-and nuclear factor kappa B(NF-κB) by traditional medicine fomular.Methods: Extracted peripheral marrow mononuclear cells from 30 human leukemic bone marrow by Ficoll and selected for CD34~+CD38~-Leukemic Stem Cells(LSCs) by MACS,every sample includes 4 groups :YuanFang fomulary,FuZHeng fomulary,QuXie fomulary and control group . To observe the effect of LSCs by MTT,and the expression of NF–κB by electrophoretic mobility shift assay(EMSA),with the traditional medicine fomular.RESULTS: Generally,the Leukemic Stem Cells were more sensitive to the YuanFang fomular than to the FuZHeng fomular and the QuXie fomular (P<0.05) with inhibition rates of 36.660±0.018%,15.230±0.043%,12.927±0.300%,respectively.And inhibiting the the expression of NF–κB is more obvious to the YuanFang fomular than to the FuZHeng fomular and the QuXie fomular (P<0.05).CONCLUSIONS: The Leukemic Stem Cells are more sensitive to the YuanFang fomular than to the other tested drugs in this study. YuanFang fomular effects the expression of NF–κB,and NF–κB is relatived with the cell apoptosis .the study offers one proof to cure AML.
