Neurodegenerative diseases refers to pathological change or barrier disease taken place in the organization of nervous system's; The psychiosis is caused by the brain dysfunction and pathological changes which can lead to the abnormality of sensation,memory,thinking,emotion and behavior. According to the World Health Organization statistics,the morbidity of neurodegenerative diseases(including aging and Parkinson's disease) and psychiosis(depression) have been increasing year by year,which is a great threat to human's health and life,and seriously affect the quality of people's lives,but also bringing great deal of loss to the whole socioeconomic. At present, there are millions of people suffering a lot from parkinson's disease,and the numbers of patient have been increasing year by year,and there is a trend that the average age of patient is decresing to younger people. Statistics indicates that the incidence rate of parkinson's disease is 1% among those above 65 years old person;and the incidence rate will be up to 2% among those above 80 years old person. The latest investigation of the University of California in the United States,also show that:global prevalence rate of depression have reached as high as 6%-11%,which has already become the world's fourth largest diseases. Along with social population aging,this have been paid widely attention from international society. These diseases have caused great losses to the family and society. If we don't take any effective measures,the losses will continue to increase. Therefore,how to approach the pathogenesis of diseases,study the effective drug treatment,improved the quality of the patient's life and reduce social burden,have already becomes one of most important topics in world medical field.At present,the cause of aging,parkinson's disease and depression are not entirely clear,and the notices are accepted that enviromental factors,genetic mutation,mitochondrial dysfunction,the excitement of the toxic damage,oxidative stress,inflammatory reaction and other factors may participate in the disease pathogenesis. In recent years,more attention have been paid to oxidative stress in the incidence of aging,parkinson's disease,depression's role in the process. Research shows that they may have the same pathogenesis,which oxidative stress may be a common feature. In neurodegenerative diseases and psychiosis,increased free radical production, lipid peroxidation,calcium homeostasis disorders as well as the release of cytochrome C,oxidative stress is the main reason. Although oxidative stress is considered that with apoptosis and neuronal cell death related,but the process leading to cell death extracellular signal receptor and its downstream signal transduction pathway is not very clear. Therefore approach the correlation of oxidative stress and nevre(psychic) disease,reveal the molecular mechanisms of oxidative stress'damage,have tremendous value for effective prevention,treatment and eventually overcome them.In this study,the aging mice models were established with abdominal cavity injection D-galactose,the hemi-parkinsionian rat models were established with striatum localization injection 6 - hydroxydopamine,the depression rat models were established with chronic mild unpredictability stress and solitary custody for the study,the relationships between aging,parkinson's disease,depression and oxidative stress and the possible role of some molecular were measured with immunohistochemistry, immunological and biochemical techniques. Our research shows that D-galactose intraperitoneal injection can decrease the locomotory capacity,spleen index,thymus index,splenic lymphocyte proliferation; 6 - hydroxydopamine into the striatum can decrease the numbers of neuron in the substantia nigra;chronic stress stimulation ,found that rats activities were decreased,the loss of interest, lack of pleasure,the numbers of neuron in the hippocampus were decreased,the expressions of BDNF in the hippocampus were decreased,these results show that the aging mice models,parkinson's disease rat models,depression rat models have been successfully established. Meanwhile,the study also found that SOD,MDA and other indicators of oxidative stress,can aggravate the oxidative stress in the body,the levels of SOD were decrased and the levels of MDA were increased,which indicate that oxidative stress participate in the process,it is the same pathogenesis. These results further shows that despite three diseases have their pathogenesis,but oxidative stress have extremely close relationship with the occurrence and development of the three diseases,which may be a major factor of causing neuronal injury. Therefore aims at the oxidative stress development corresponding medicine,possibly displays the extremely good effect in these three kind of disease's treatment.
