IntroductionAtopic Dermatitis(AD)is a chronic inflammatory skin disease and is concerned with heredity,its pathogenesis and clinical manifestation are complicated.Filaggrin is a key protein involved in skin barrier function.Recently,mutations in the filaggrin gene, FLG,were identified in European families with ichtyosis vulgaris(IV) and shown to be an important predisposing factor for atopic dermatitis(AD).Study on the relationship between OSAAD(objective severity assessment of atopic dermatitis) of skin barrier function and SCORAD score system.A commonly used clinical measure for AD today, SCORAD,relies on subject critereia in assessing the severity of lesions and the extent of involvement.OSAAD assessing AD severity objectively by measureing skin barrier function,and by utilizing Noninvasive bioengineering methods and computer-assisted estimates,all of these instruments are non-invasive.We will study the skin barrier function depended on measureing epidermal water capacitance,sebum,and PH values. The objective scoring system avoids much of the researchers' subjectivity.ObjectiveTo study the role of FLG polymorphisms in skin barrier function of AD in Chinese,and measure skin water capacitance,sebum secretion,and PH values in 32 cases AD patients and 30 cases healthy normal control subjucts with non-invasive measurement,and analyze their relationship with skin barrier function.Methods59 AD patients and 169 control subjucts in Liaoning aged from 6 months to 21 years old were analyzed for the presence of FLG E2422X,S2554X,S2889X,S3296X, and R4307X mutations,by using polymerase chain reaction(PCR) approach.The skin water capacitance,sebum,PH values,in the forehead,cubital fossa,back of hand, breast bone front,sacro-waist,and shin front for 32 cases AD patients and 30 cases healthy normal control subjucts in Liaoning aged from 6 months to 21 years old were measured using the Soft 5.5 ITALIA CALLAGARI S.P.A.Via Adamello,2/A,Divided the 32 cases AD patients into 2 groups,mildly affected,and severely affected areas, according to different slowed pulses,including dry,erythema,desquamation,erythema,papula and effusion.ResultsThe test results revealed that all the mutations were not present in AD patients and control subjucts.The epidermal water capacitance,sebum,and PH values of measured anatomic-sites and different slowed pulses were different.The biophysical parameters between normal skin of AD patients skin and healthy individuals were significant correlations(p<0.05),the biophysical parameters between mildly affected,and severely affected areas of AD patients skin were significant correlations(p<0.05). Water capacitance and sebum in forehead,cubital fossa,and breast bone front were statistically positively relevant(r=1.830 r=1.057 r=0.294 P<0.05),but the skin PH values and water capacitance in breast bone front,sacro-waist,and shin front were statistically negatively relevant(r=-17.792 r=-10.683 r=-14.143 P<0.05).ConclusionThe results may be effected by little sample size,or regions and races. Non-invasive and objective evaluation can explore the changes of skin water capacitance,sebum,PH values in different condition and different slowed pulses,the biophysical parameters reveal their significant relevance.These parameters were sensitive to reflect the skin barrier function and severity extent of disease.
