Study On The Quality Control And Pharmacodynamics Of Nongbai Mixture

ObjectiveTo improve the quality criteria of Nongbai mixture.To approach the effects of stopping coughing,eliminating phlegm,inhibiting asthma and anti-inflammation in Nongbai Mixture and simplified formula.To observe the acute toxic reaction and death of mice one day after intragastric administration of Nongbai Mixture and simplified formula so as to appraise their safety.To investigate the internal mechanism on treating chronic bronchitis of Nongbai Mixture.MethodsImprovement On Quality Criteria:Thin-layer chromatography(TLC) is used make qualitative determination of Semen Trichosanthis,Radix Peucedani,Fructus Perillae and Reverse Phase-High performance liquid chromatography(RP-HPLC) is used to determine the content of its main constituents of Ephedrine Hydrochloride and Pseudoephedrine Hydrochloride,berberine hydrochloride,baicalin.Pharmacodynamic Test:Use the methods of ammonium hydroxide in mice,citric acid in guinea pigs,trache-phenol red excretory model in mice,histamine phosphate in guinea pigs,auricle edema induced by xylene in mice and paw edema induced by carrageenin in rats to approach the effects of stopping coughing,eliminating phlegm, inhibiting asthma and anti-inflammation in Nongbai Mixture and simplified formula. The mice were intragastrically administered with Nongbai Mixture and simplified formula to observe the acute toxic reaction.Mechanism of Action:Duplicated the animal model of chronic bronchitis with refined smoking-fumigated method,determined the contents of TNF and ET in the serum and lung tissue.ResultsOne.Semen Trichosanthis,Radix Peucedani,Fructus Perillae can be identified by TLC.The contents of berberine Ephedrine Hydrochloride and Pseudoephedrine Hydrochloride,hydrochloride,baicalinby RP-HPLC are stable and accurate.The recovery of Ephedrine Hydrochloride is 98.28%and RSD is 0.94%;the recovery of Pseudoephedrine Hydrochloride is 98.01%and RSD is 0.71%;the average recovery of berberine hydrochloride is 98.40%and RSD is 1.53%;the average recovery of baicalin is 98.83%and RSD is 1.82%.Two.Nongbai Mixture and simplified formula can prolong latent period of cough in mice and guinea pigs and decrease the frequency of cough,increase phlegm-eliminating quantity of phenol red in mice,resist the symptoms of asthma by histamine phosphate in guinea pigs and prolong the latent period of falling down,and lessen the auricular edema induced by xylene in mice and paw edema induced by carrageenin in rats.The maximum dosage of Nongbai Mixture is 170.4g·kg^-1 and the maximum dosage of simplified formula is 80.0g·kg^-1,equivalent to men's daily administration dosage by 160 times.Three.The contents of TNF and ET in the serum and lung tissue of the model group were significantly improved than that of the normal group(P＜0.01).Compared with the model group,the contents of TNF and ET in the serum and lung tissue of each treating group were significantly reduced(P＜0.05 or P＜0.01).ConclusionsOne.The present method is simple,accurate,sensitive and highly reproductive, which can be used for the quality control of this preparation.Two.Nongbai Mixture and simplified formula had good effects of stopping coughing,eliminating phlegm,inhibiting asthma and anti-inflammation.Nongbai Mixture and simplified formula are relatively safe.Three.Nongbai Mixture can control chronic inflammatory reaction by reducing the contents of TNF and ET.
