ObjectiveTo investigate the growth and apoptosis of Decoction of cooling blood for psoriasisOn Malpighian cell lines Hacat and their cell cycles involved.MethodsThe effect of Decoction of cooling blood for psoriasis on the grow th of Hacat were studied through M TT assay. The effect of Decoction of cooling blood for psoriasis on the apoptosis of Hacat were studied by combined Annexin-V protein iodide staining. The effect of Decoction of cooling blood for psoriasis on the cell cycle of Hacat were studied by a propidium iodide method.ResultsDecoction of cooling blood for psoriasis inhibited the Hacat cells in a time-manner. T reatment with 0. 4—10mg/ml Decoction of cool ing blood for psoriasis for 48h induced apoptosis of Hacat cells and the apoptosis rate reached (31.77±5.88) % at concentration of 10mg/ml. Flow cytometric analyses showed that Hacat cells treated with Decoction of cooling blood for psoriasis were accumulated inG1 phase in the cell cycle, the ratio of cells in andS phase wasdecreased at 48 h treatment.ConclusionDecoction of cooling blood for psoriasis is able to inhibit theproliferation of Hacat cells by regulating the cell cycle andinducing the cell apoptosis.
