Objectives: To investigate the effects of alprostadil and norepinephrine and anisodamine on model of septic shock rat,by comparing induced nitricoxide synthase(iNOS) level in nephrid- ial tissue and endothelin in serum and renal cell apoptosis, to evaluate the roles of all those medicines on renal microcirculati- on and renal function.Methods:Fifty male Wistar rats weight 200-250g were randomly divided into five groups,each group includes ten rats. control group(groupâ… ),septic shock group (groupâ…¡) ,treatment group with norepinephrine (groupâ…¢),treatment group with pro- stadil(groupâ…£),treatment group with anisodamine (groupâ…¤), Each group was divided into 1h and 3h subgroups (n=5 in each subgroup).Septic shock model was made by injecting lipopolys- accharide(LPS) via left femoral vein continuedly. Groupâ… was given normal sodium also via left femoral vein continuedly;â… tube was establish in left carotid artery in each rat, blood pressure can be measured by using those tubes. Incision of trachea was also employed in all rats. In groupâ…¡,LPS was injeted via left femoral vein by the rate 10mg/kg,norepinephrine was injected via left femoral vein by the rate of 50ug/kg.h in groupâ…¢; alprostadil was injected by the rate of 0.1mg/kg.h in groupâ…£; anisodamine was injected via left femoral vein by the rate of 0.1mg/kg.h in groupâ…¤.groupâ… and groupâ…¡were injected equal volume by equal rate.all rats were killed after 1h and 3h.serum was taken out and used to measure endothelin(ET)by means of ELISA method; renal was homogenate after velamen and renal pedicle were removed,then homogenation was used to measure iNOS and renal cell apoptosis.Results:1 iNOS in nephridial tissue in groupâ…¡and groupâ…¢were heightened, which was also found heightened in 1h in groupâ…£and groupâ…¤,but degraded in 3h, difference was significant. Compared with groupâ…¢and groupâ…¡, iNOS in groupâ…£and groupâ…¤were lower, difference was significant.2 Renal cell apoptosis level in nephridial tissue in groupâ…¡and groupâ…¢were found heightened gradually in 1h and 3h, which were strengthened in groupâ…£and groupâ…¤, difference was significant. The difference between groupâ…¡and groupâ…¢was not significant.3 Endothelin(ET)level in serum the groupâ…¡was found heightened gradually with the time prolonged, the difference between groupâ… and groupâ…¡was significant; groupâ…£and groupâ…¤compared to groupâ…¡the difference was significant, the difference between groupâ…£and groupâ…¤w as not significant. Conclusions:1 A septic shock model can be made successfully by injecting LPS via vein.2 Anisodamine and alprostadil all can improve renal function and the induced nitricoxide synthase and endothelin(ET),renal cell apoptosis were found heightened gradually in septic shock.3 Anisodamine and alprostadil can lower iNOS level and renal cell apoptosis in nephridial tissue, maybe owing to rival the effects of ET.PGEI can rival the Ca2+ in vascular smooth muscle cell, and then can rival the sympathnetic nerve ending to delievr norepinephrine.Anisodamine and alprostadil all can improve microcirculation.4 There is seemly no obvious effects of norepinephrine on improving renal function,it also seems no obvious effects on lessening iNOS level in nephridial tissue maybe in early phase of shock. ET plays an important role, it contracts blood vessel and norepinephrine can not improve organ perfusion.
