Resveratrol is found to pose many specific pharmacological functions such as anti-fungal, anti-bacterial,anti-virus anti-atherosis,antineoplastic,anti-oxidation,reducing the risk of heart brain blood vessel disease(CHD),et al.Resveratrol was extracted mainly from limited Chinese medicine Polygonum cuspidatum at present.Therefore,it is meaningful to develop other new material for extracting resveratrol in view of environmental protection and economic value.The abandoned peanut plants after harvest are selected to used as raw materials in this thesis.To optimize the extraction technique for resveratrol from peanut plants by alcohol refluxing extraction,the single factor tests and response surface method were used and purification of resveratrol by column chromatography was utilized.High purity of resveratrol was gained by recrystallization,thus a new green technology of protecing environment is promised to be developed to exetract and purify resveratrol from peanut plants.(1) HPLC analysis sample resveratrol were adopted as effective method in this paper.The extracts were analyzed analysis of Kromasil(4.6mm×250mm,5μm) column,column temperature was 25℃,using acetonltrile-water(30:70) system as the mobile phase,and the the flow rate was 1.0 mL/min,detection wavelength was 305 nm,the injection volume was 20μL.(2) Refluxing extraction was used for extracting resveratrol from a kind of new raw material named as waste peanut plants.On the basis of single factor analysis,with the extraction percent and purity percent as evaluation index,the optimal conditions to obtain extraction percent and purity percent were determined by three-level three-factor Box-Behnken design.The results showed that the optimum extraction conditions were:the volume fraction of ethanol was 76.27%,extracting time was 3.22h,solid-liquid ratio was 1:11.01(g:mL), extracting times were:3.The extraction rate and purity of resveratrol were 87.07%and 0.069% respectivel-y,and it s desirability was 0.900.(3) Based on the research of separation conditions,the optimum conditions of resveratrol were:adding distilled water with 6 times volume,acetic ether-water(1:1 v/v),extracting times were:3,extractum was obtained by recovering ethyl acetate,petroleum ether- extractum(6:1 v/w).The results showed that the extraction rate and purity of resveratrol were 86.41%and 2.13%respectively(4) Silica gel column chromatography was used in the separation and purification of resveratrol,and the optimum conditions were as fellows:dry-process was selected as sampleloading method,samples:silica(1:50),eluant of column chromatogaraphy was acetone-nhexane (3:7).The purity and yield of resveratrol were 71.35%and 93.49%respectively. Methylene chloride-ethylacetate(5:1,v/v) and trichloromethane-ethylacetate(5:1,v/v) were used to purify the rude resveratrol to the purity and yield reaching 98.01%and 90.59%by cystallization and re-cystallization respectively.After the resveratrol was purified and detected by HPLC from the sample and criterion figure,we know that peak model was the same basically,and the absolute contents of resveratrol was 98.23%.(5) In order to transform agriculture solid waste to products with high economic value and practical value,a new process and technology of environment-friendly "green-tech" is developed to extract resveratrol from the waste peanut plants.
