| Resveratrol belongs to two styrenecompounds, mainly distributed in Polygonum cuspidatum Sieb et Zucc, grapes, peanuts and other plants, the earliest found as a Vitis plant antitoxin. Domestic and foreign scholars have done the in-depth study on the resveratrol, they found that the material not only has an antibacterial effect, there are significant antioxidant, anticancer pharmacological effects of resveratrol, which promote huge development of Resveratrol in the pharmaceutical industry, in addition, resveratrol are also widely applied in food, chemical, cosmetics industry. In this paper, taking Polygonum cuspidatum Sieb et Zucc as raw material, the extraction process of Resveratrol from Polygonum cuspidatum Sieb et Zucc and purification were studied, the extraction and analysis method,andisolation and purification conditions are confirmed. The specific contents are as follows:1. Determine determination and analytical methods of Resveratrol.Based on ACN-WATER in equal elution, determine the content of resveratrol in Polygonum cuspidatum Sieb et Zucc extract by HPLC. By comparing various expansion system, the TLC was also established,that provides qualitative determination method for subsequent experments.2. Determine extraction conditions of Resveratrol from Polygonum cuspidatum Sieb et Zucc. Carrying out extraction by water bath reflux method, through the single factor test, checking out effect of solvent concentration,solvent consumption, extraction temperature,extraction time and extraction times on Resveratrol content in Polygonum cuspidatum Sieb et Zucc. Base on this result, to optimize the extraction process of Resveratrol from Polygonum cuspidatum Sieb et Zucc by using response surface method, the optimal conditions of the extraction were determined:extracted two times in12.7times the amount of72%ethanol solution50℃water bath reflux, each time1.1h.The average content of resveratrol was2.80mg·g-1, close to the predicted values of2.82mg·g-1.3. Study on isolation and purification of Resveratrol. Through the static experiment, the suitable adsorption resin was chosen. The adsorption and desorption properties of resin were studied through the dynamic test, the best adsorption and elution conditionswere determined. Resveratrol was concentrated, finally silica gel-column chromatography should be used for further purification. By HPLC detection, obtaining the product of the89.03%purity. |
