Trans-Resveratrol is a polyphenol widespread in grapes, peanuts and Polygonum Cuspidatem as the major active compound of phytoalexin. It has many biological and pharmacological activities, such as anti-cancer, anti-microbial, anti-oxidant and estrogenic activity. It also can prevent cardiovascular diseases. Because of its valuable uses in healthcare, people has focused on its extraction and purification from plant resources in recent years.Extraction and purification of active components from natural products relate to selecting solvents and determining the ratio of mixed solvents. It is important to study the solid-liquid equilibrium of natural active component in different solvent systems for optimizing technology.In the first chapter of this thesis, we mainly introduced the study history, physicochemical properties, pharmacological effects, biological sources and synthetic methods of trans-resveratrol. On this basis, the research contents of this thesis were described.In the second chapter, a simple procedure to analyze trans-resveratrol with HPLC method was set up. This method was used to study the light-stability of trans-resveratrol. Through comparison of eluting capability with various ratios of acetone and water, the optimum extraction condition with macroporous resin was found. In this thesis trans-resveratrol was purified by recrystallization with ethanol and water mixtures, and an orthogonal design had been taken to optimize the condition of recrystallization including the ratio and volume of ethanol and water mixtures, likewise the number of recrystallization.In the third chapter, the solid-liquid equilibrium of trans-resveratrol was studied through equilibrium method. The solubilities of trans-resveratrol in eleven pure solvent, ethanol + water and acetone + water mixed solvents were measured using HPLC analytic method at different temperatures.In the forth chapter, a three-parameter equation was used to correlate the experimental data of trans-resveratrol solubilities in different solvent systems and showed a satisfactory results. Then NRTL equation and UNIQUAC equation were also used to correlate the experiment data of binary systems, and we got interaction parameters between solute and solvents. As a result, UNIQUAC activity coefficient model was better than NRTL activity coefficient model in binary systems. Moreover, in ethanol + water and acetone + water ternary systems, UNIQUAC equation was used to correlate the experimental data to obtain interaction parameters. We compared the difference between experimental values and calculated values and discussed the feasibility of UNIQUAC equation to correlate the solubilities of trans-resveratrol in ternary systems.In the last chapter, all of the experimental work was summarized. We found the deficiencies in this thesis and proposed the idea of latter research.
