Objective:To investigate the expression of MCP-1 at mRNA and protein levels in serum and /or cerebrospinal fluid(CSF) of patients with Intractable Epilepsy and explore its role in the onset.Methods:The patients were divided into 3 groups:Intractable epilepsy (Group A),non-intractable epilepsy group(Group B) and control group(Group C),and the number of each group is 30,32,32.The mRNA and protein expressions of MCP-1 in serum and/or CSF were detected by semi-quantitative reverse transcription- polymerase chain reaction(RT-PCR) and enzyme-linked immunosorbentassay(ELISA),and compared to each group' result.Results:1.The protein expressions of MCP-1 in serum and cerebrospinal fluid detected by ELISA.The expressions of MCP-1 were 133.29±22.361 (serum) /143.05±17.336(CSF) in Intractable epilepsy group,54.12±7.644 (serum)/ 28.75±3.975(CSF) in non-intractable epilepsy group,and 51.19±6.547(serum) in control group.The expressions of MCP-1 in serum of intractable epilepsy group was significantly higher than those of control group and non-intractable epilepsy group(P<0.01 ).The expressions of MCP-1 in CSF of intractable epilepsy group was also higher than those of non-intractable epilepsy group(P<0.01),but the expressions of MCP-1 in serum of non-intractable epilepsy group were not higher than that in control group (P>0.05).2,The mRNA expressions of MCP-1 in the three groups were all positive. By semi-quantitative RT-PCR,we detected that the mean relative explession of MCP-1 mRNA in the peripheral blood of intractable epilepsy group 0.855±0.054,and that in non-intractable epilepsy group and control group were respectively 0.524±0.039 and 0.515±0.036.Analyed by one-way ANONA,it meant that the expressions of MCP-1 mRNA in the peripheral blood of intractable epilepsy group was much higher than that of the patients in control group and non-intractable epilepsy group,and the difference was of significance(P<0.01).Otherwise,the difference between non-intractable epilepsy group and control group were of no significance(P>0.05).Conclusions:The mRNA and protein expressions of MCP-1 is significantly increased in serum and CSF of the patients with intractable epilepsy.The inflammation and immunization factors that MCP-1 participated maybe play important roles in the onset process of Intractable epilepsy.
