Objective:To explore the relationship between the level of serum sHLA-G , HLA-G14bp insertion / deletion polymorphism and recurrent spontaneous abortions through the mensuration of the level of sHLA-G and HLA-G14bp insertion / deletion genotype of the normal pregnancy women and the one with recurrent spontaneous abortions .Methods: The 14bp insertion / deletion polymorphism of HLA-G gene in 60 recurrent spontaneous abortions patients (case group) and 60 the normal pregnancy women(control group) was detected by polymerase chain reaction-restrictive fragment length polymorphism(PCR-RFLP) ; The level of serum sHLA-G was detected by Enzyme-linked immunosorbent assay(ELISA). Group-samples T test was used to compare the level of serum sHLA-G between case group and control group.χ2 analyses were used to compare genotypic frequencies between two groups.( because 14 bp insertion homozygous and 14bp heterozygous deletions was genotypes for the sHLA-G low secretion , so together statistics.) Analysis of variance(LSD-T test) was used to compared the level of sHLA-G among different genotypes.Results:(1)The levels of sHLA-G in serum of the case group were significantly lower than that of in the control group(t=2.583, p <0.01). the difference was statistically significant;(2)the genotype frequencies of +14bp/+14bp homozygous and +14bp/-14bp of the heterozygous in the case group were significantly higher than that of the control group(χ2=7.28 , p <0.05). the difference was statistically significant;(3)the level of sHLA-G in serum with +14bp/+14bp and +14bp/-14bp genotype were significantly lowerer than the corresponding with -14bp/-14bp genotype(p <0.01).the difference was statistically significant.Conclusion:The low level of sHLA-G in serum and homozygous (+14bp/ +14bp) and heterozygous (+14bp/ -14bp) genotype is the risk factors of recurrent spontaneous abortions; The level of sHLA-G in serum may be corrected to HLA-G14bp insertion / deletion polymorphism.
