| BackgroundSpontaneous abortion(SA) is one of the common complications during pregnancy,however the mechanism of SA has not been completely understood.The change of extracelluar matrix (ECM) composition has a close relationship with SA,and heparanase can have an effect on pregnancy by analyzing the ECM.The balance between heparanase and heparin is important for the body and is relevant to SA.ObjectiveTo observe the potential pathologic significants of heparase in placenta in the course of spontaneous abortion rats with bromocriptin,and the interfering effect of the treatment of low molecular weight heparin(LMWH). Methods10-12w-year-old wistar female rats were fed with male rats in one cage,with the proportion of 2:1.The sign of pregnancy is finding sperms in the vaginal paint,which was regarded as the first day of pregnancy.All of the pregnant rats were randomly divided into three groups,that was normal pregnant group(A group,n=20),spontaneous abortion group with hypodermic injection of bromocriptin 0.3mg/kg/d during the day of 3---8(B group,n=20),and LMWH interfering group with hypodermic injection of LMWH 100IU/kg/d during the day of 8---12 after bromocruptin(C group,n=20).At the time points of the 13th day we had Caesarean operation on all rats and detected the protein expression of heparanase in placenta by immunohistochemical ways.All the data of experiment were analyzed by statistics software SPSS13.0.Results1 Abortion rate and embryo number:(1)Group A:abortion rate was 0.11 and pregnant rate was 0.89 and embryo number was 10.05;Group B:abortion rate was 0.72 and pregnant rate was 0.28 and embryo number was 4.65;Group C:abortion rate was 0.47 and pregnant rate was 0.53 and embryo number was 6.65.The pregnant rate and embryo number of group A and C was significantly higher than group B(P＜0.05).2 Expression of heparase proteins:The heparase protein was expressed in all decidual tissue in all groups.(1) The expression area of heparnase in spontaneous abortion group was 22108.22±514.66,and was higher than normal pregnant group(2487.55±194.17).There was a significant difference between the two groups.(2)The expression area of heparanase in LMWH interfering group was 19027.55±1994.61,and had a great difference with spontaneous abortion group.(3)There was remarkable difference between normal group and LMWH interfering group.Conclution1 In the course of spontaneous abortion in rats,the up-regulated expression of heparanase exhibit a great malgenic sense,while the LMWH treatment have a visible interfering effects.
