Lotus leaf is a sort of crude plant material for medicine and food.Polyphenols have health functions,such as antioxidation,fighting against senium,curing cardiovascular diseases,reducing blood fat and so on.we use the lotus leaf as raw materials,the best extraction process were researched and the HPLC fingerprint of flavonoids from different medicinal Lotus Leaves was established.And we will identify the polyphenol by the color reactivity:1.Microwave extraction:the power of microwave low,the period of microwave treatment 150 seconds,20%ethanol,liquid material ratio1:60.The rate of extracting of polyphenols in lotus leaf was 11.80%under this condition.2.Ultrasonic extraction:The optimal process conditions were percent of ethanol volume 60%,ratio of solid to liquid 1:14(g/mL),four extraction times,and extraction period as 15 min.The rate of extracting of polyphenols in lotus leaf was 4.85%under this condition.3.Establish the fingerprint by study the HPLC fingerprints of polyphenols from different medicinal Lotus Leaves,establish the detection of the HPLC fingerprints of Lotus Leaves by comparing the Chromatography conditions,such asmobile phase, wavelength and flow speed,the analytical results of methodologic experiments were favorable.Calibration 12 common peaks in the fingerprint of polyphenols from 10 Variety Lotus Leaves,the area of commen peaks is above 90%in the area of all peaks.All uncommen peaks were in the lower and unstable in samples.4.The study of anti-oxidative character of polyphenols in lotus leaf.The deoxidize ability,eliminative ability of O2-·and·OH,anti-oxidative ability were good.
