Objective: Through the detection of Survivin in normal cervical tissue, CIN, cervical cancer and lymph node metastasis without lymph node metastasis specimens and samples of the expression of Survivin with further study of the pathological types of cervical cancer, clinical stage, the level of credits, as well as lymph node the relationship between the transfer to explore the incidence of cervical cancer, the development of mechanisms for the early diagnosis of cervical cancer and provide a theoretical basis for gene therapy. Methods: 130 cases of specimens from March 2005 to March 2008 in the Third Clinical Medical College of Jilin University (China-Japan Friendship Hospital) and the Third Clinical Hospital of Jilin University of obstetrics and two out-patient clinics and hospitalized patients of cervical tissue wax block. Preoperative patients were not receiving any treatment. Divided into three groups: (1) normal control group: benign uterine diseases because of hysterectomy, postoperative pathology confirmed 22 cases of normal cervix. Aged between 29-56 years old, average age 41.6 years old; (2) CIN groups: in the parallel out-patient treatment of cervical cone excision, 44 cases confirmed to be pathological CIN, of which 21 cases of CINⅠ-Ⅱand 23 cases of CINⅢ, aged between 26-60 years old, average age 43.8 years; (3) cervical cancer group: 64 cases of cervical cancer, aged 26-62 years old, average age 44.2 years, of which 56 cases of squamous cell carcinoma, adenocarcinoma 8 patients, preoperative biopsy had cervical pathology. Reference to malignant tumor specimens according to FIGO staging in 2000,Ⅰperiod of 28 cases,Ⅱperiod of 36 cases, both the implementation of extensive hysterectomy and pelvic lymphadenectomy. Pathological examination of return: one with 37 cases of lymph node metastasis; histopathological grade (WHO classification criteria in accordance with the histological grade): 34 cases in which poorly differentiated, 30 cases, the well-differentiated. The average age of more than three sets of comparison there was no significant difference(P<0.05), comparable. Three groups of cervical tissue paraffin blocks, serial sections 4μm thick, prepared by each of the three cases, after the back-up dry. For HE staining, respectively, Survivin immunohistochemical staining, using the known breast cancer-positive specimens were positive for Survivin to PBS instead of primary antibody for negative control. All sections were blinded experts identified pathology. By immunohistochemical SP method, are equipped with negative and positive control to observe the positive use of double-blind method. Survivin immunohistochemical staining mainly in cytoplasm appeared granular brown its positive criteria. According to the percentage of positive cells divided into: negative (-): positive cells <1%; weakly positive (+): 1% positive cells -10%; moderately positive (+ +): 11% positive cells - 50%; strong positive (+ + +): positive cell count> 51%. Count data usingχ~2 test. All statistics were carried out in SPSS11.5 software. To P<0.05 for statistical significance. The results:①Survivin in the 22 cases of normal cervical tissue positive expression rate of 0%, 21 cases of CINⅠ-Ⅱtissue in 6 cases Survivin positive expression rate was 28.6%, 23 cases of CINⅢorganizations Survivin in the 14 cases positive expression rate was 60.9%, 64 cases of cervical cancer in 55 cases of Survivin positive expression rate of 85.9 percent. Cervical cancer rate of positive expression of Survivin in CINⅢtissues Survivin positive expression rate compared with a significant difference(P<0.05). CINⅢtissue expression of Survivin-positive rates of CINⅠ-Ⅱtissue the rate of positive expression of Survivin, compared with significant difference(P<0.05). CINⅠ-Ⅱorganizations Survivin positive expression rate of normal cervical tissue with positive expression rate of Survivin comparison, a significant difference(P<0.05). Description Survivin expression in cervical carcinoma and cervical cancer occurrence and development of the process is closely associated with the occurrence of cervical cancer, development, Survivin expression was the trend continued.②Survivin in the cancer group of stageⅠpositive expression rate of 71.4% (20/28), II Phase group positive expression rate of 97.2% (35/36), Survivin in cervical cancer group II high rate of positive expression I view the group in the positive expression rate, the difference has a significant (P<0.05). Survivin in cancer cells that the expression of plasma intensity and clinical stage of cervical cancer, with an increase in clinical stage, cervical cancer organizations continue to enhance the expression of Survivin.③with lymph node metastasis in 37 cases of cervical cancer organizations, including 36 cases of Survivin positive expression rate was 97.3%, without lymph node metastasis in 27 cases of cervical cancer organizations, including 17 cases of Survivin positive expression rate of 70.4%. With lymph node metastasis of cervical carcinoma Survivin positive expression was higher than that without lymph node metastasis of cervical carcinoma The positive expression rate of Survivin, a significant difference (P<0.05). Description Survivin expression in cervical carcinoma and cervical cancer cases occurred in the transfer of any closely related to each other.④56 cases of cervical squamous cell carcinoma in 48 patients with Survivin positive expression rate was 85.7%; 8 cases of cervical adenocarcinoma in seven cases of Survivin positive expression rate of 87.5 percent, the second group there was no significant difference (P<0.05). That cervical cancer tissue expression of Survivin and non-pathological types of cervical cancer related to each other.⑤high in cancer, Survivin differentiated group of the positive expression rate of 73.3% (22/30), poorly differentiated group Survivin positive expression rate was 97.1% (33/34). With the lower degree of pathological differentiation, Survivin positive expression rate gradually increased, and a significant difference(P<0.05). Description Survivin expression in cervical carcinoma with cervical cancer have a pathological degree of differentiation is closely related to sexual.⑥Expression of Survivin-positive cells are mainly located in cytoplasm colored in brown were. Conclusion:1. Survivin in normal cervical tissues without expression in CINⅠ-Ⅱtissue expression of a relatively low level, in the CINⅢexpression of middle level organization, in the high-level expression in cervical carcinoma, suggesting that Survivin in cancer occurrence and development plays an important The role of.2. Survivin in the cancer tissues with positive expression rate of the pathological types of cervical cancer has nothing to do. And later clinical stages of cervical cancer, the degree of histopathological differentiation of the lower cervical carcinoma Survivin positive expression rate of the higher. Description Survivin expression and clinical stage of cervical cancer, the degree of pathological differentiation, with the development of cervical cancer, cervical cancer organizations continue to enhance the expression of Survivin. With lymph node metastasis of cervical carcinoma The positive expression rate of Survivin was significantly higher than that without lymph node metastasis of cervical carcinoma The positive expression rate of Survivin that Survivin and cervical cancer with lymph node metastasis is closely related to sexual.3. Cervical cancer by detecting the expression of Survivin to determine the degree of malignancy of cervical cancer has important significance and prognosis. Monitoring of cervical intraepithelial neoplasia in the expression of Survivin is expected to become cervical precancerous lesions predict whether an indicator of progress.4. Survivin as a tumor-specific apoptosis inhibitory genes, gene therapy can be used as the target gene, its cervical cancer treatment in a wide range of applications.
