PurposeCell apoptosis plays an important part in many human malignant tumor pathogenesis. Cells apoptosis is adjusted mutually by apoptosis inhibiting genes and apoptosis activating genes. Cell apoptosis is regulated by both. Survivin is a member of the inhibitors family of apoptosis. It is a profilin of apoptosis with very strong activity. Survivin inhibits caspase activity through direct binding with the cysteine protease, caspase - 3 and caspase - 7, and inhibit apoptosis. The distribution of Survivin has conspicuous selectivity of cell. It almost doesnt express in normal tissues of adults, while express in all tumor studied by human, and the difference is only the expressing level. Survivin expresses in all gynecol-ogy malignant tumors, for instance, ovarian epithelial cancer, endometral cancer, uterine cervix cancer and so on. Fas is a signal molecule of cell apoptosis. Fas can transmit death signal when binding with FasL, and lead to the death of the target cells expressing Fas. Both Survivin and Fas affect apoptosis program and play important parts. In the article we try to study the expression and the relation between the inhibition protein " Survivin" and the apoptosis signal molecule "Fas" in normal cervix, cervical intraepithelial neoplasia, CIN and cervical carcinoma and to investigate whether both affect the development of cervix carcinoma and what parts they play.MethodsUsing strptvidin -biotin peroxidase (sp) method, we examined the proteinexpression of Survivin and Fas in 9 normal cervixes, 15 cervical intraepithelial neoplasias, CIN and 34 cervical squamous carcinomas tissues. All specimens, which had been conformed by pathology, were collected through biopsy and operation from the china medical university NO. 1 accessary hospital and the lia-oning province tumor hospital from 2000 to 2002 year, and were reviewed by the same pathologist.Results(1) The positive expression rates of Survivin and Fas were 0 88. 89% in normal cervical tissues, 40% 53.33% in cervical intraepithelial neoplasia tissues and 67. 67% 44. 12% in cervical squamous carcinomas tissues respectively. There were significant differences in the positive expression of Survivin and Fas in different cervical diseases (P <0.05). The expression of Survivin tended to increase, while Fas tended to decrease with the development of tumor in difference cervical diseases. (2) The positive expression of Survivin was associated with higher tumor grade and clinical stage ( P < 0. 05 ). The later was the clinical stage and the higher was the pathologic grade of uterine cervix cancer, the stronger was the expressing of Survivin. The positive expression of Fas was only associated with tumor grade ( P < 0. 05 ). The higher was the pathologic grade of uterine cervix cancer, the poorer was the expressing of Fas. (3) The correlation analysis and statistics analysis of expressing of Survivin and Fas protein and the clinical pathological grade of uterine cervix cancer indicated that there were statistical significance in difference of Survivin and Fas expression, and positive expression of them was negatively relative (P <0.05).ConclusionsThe article first studied the dynamic expression of Survivin in the development of cervix cancer in the country. The result indicated that Survivin wasnt detected in normal tissue, which was uniform with the documents published, and was detected in cervical intraepithelial neoplasias by 40 percents. The resultdisplayed that the expression of Survivin can emerge in the early stage of cervix malignization. The expression of Survivin was visibly stronger in cervix cancer, the rate of the positive expression was 67. 67%. Moreover, the analysis to clinical data discovered that the later was the clinical stage and the lower was the histological differentiation, the higher was the positive rate of Survivin, which hinted that the expression of Survivin protein suggest stronger invasion and in-faust prognosis of cervix cancer.
