Purpose:The research tested 135 ordinary people and 16 Long-distance athletes of Uyghur nationality in Xiniang, Try to define the polymorphism marks of ACE gene related to excellent endurance, analysis how much ACE gene transcribe in leucocyte and how much ACE in serum. To discuss molecular mechanism of ACE gene affect aerobic endurance capability. Provide a criterion and experimental method for the athletes'gene selection.Method: Uyghur nationality people ACE gene I/D Polymorphism were tested through Polymerase Chain Reaction (PCR). Athletes people and control group ACE gene transcribe in leucocyte were measured with a reverse transcription and semi-quantification polymerase chain reaction (RT-PCR). Serum ACE was tested by enzyme kinetics method. The results were analysised by Chi-square test and One-Way ANOVA of SPSS.Result: Contrary to Han nationality, there are significant genotype and allele frequency different of Uyghur nationality (P<0.05). And there are no significant different between Uyghur nationality and European and American people (P>0.05). II genotype frequency of Long-distance athletes are noticeably different from that of control group (P<0.05), and there are no noticeably different from that of excellent athletes have been studied by home and abroad (P>0.05). There are noticeably different from that of 3 genotype on ACE mRNA transcribe of control group on condition of rest, after Exercise ACE mRNA were decline, but there are no significant different(P>0.05), and athletes group had the same trend, trancribe level:DD>ID>II. Serum ACE level is no signigicant different between athletes group and contronl group (P>0.05), but there are noticeably different between 3 genotype (P<0.01).Conclusion: The frequency distribution of ACE Gene Polymorphism of Uyghur people has very clear racial characteristic. Despite Uyghur is the race which migrations of various populations[1], we conclude from this research that it's genetic background more close to Caucasians; For the long-distance runner of Uyghur nationality, I allele and II genotype can be considered the gene marks of excellent endurance and the form of their expression has a clear racial distinction which is greatly different from the result of other nationality. Uyghur people who has genotype DD or D allele had higher ACE mRNA levels in white cell of blood and serum ACE activity, the conclusion indicate that there is negative correlation between sport ability and ACE level, so I think ACE gene Polymorphism can change its mRNA transcribe to adjust ACE translate, and then adjust Aerobic Endurance Capability of human body.
