Objective: To confect modified self-confected LPD solution; to compare the effectiveness of lung preservation between modified self-confected LPD solution and LPD solution (Perfadex) in canine left single lung transplantation model; to evaluate changes of data monitoring lung preservation after defferent periods preserved with modified self-confected LPD solution.Materials and methods: Self-confected LPD solution was confected based on public prescription of LPD solution. The modification was performed by prostaglandin E1 (125ug/L) and nitroglycerin (100ug/L) added in self-confected LPD solution. 32 healthy dogs were divided into two groups randomly: donor group 16; recipient group 16. 16 canine left single lung transplantations in situ were performed and also divided into two groups randomly: 8 of them as control group were flushed with LPD solution, the other 8 as experimental group were flushed with modified self-confected LPD solution. In vivo, changes of MPO, MDA, W/D, pathological result and super-micro structural result from donor lung tissue got at pre-flush, after flush, 4-8-12-24h of preservation and 4h after transplantation were compared between two groups, respectively. In vitro, changes of pulmonary vascular resistance, pulmonary conformance and systemic oxygen supplement of recipient after transplantation were also compared between two groups, respectively. Statistical analyses were performed by using Independent-Samples T test.Results: The physicolchemical property of modified self-confected LPD solution was close to LPD solution. No significant difference was found between two groups either from the changes of MPO, MDA, W/D, pathological result and super-micro structural result, nor from the changes of pulmonary vascular resistance, pulmonary conformance and systemic oxygen supplement (P value>0.05). But the lesson of pathological result and super-micro structural result from experimental group were slightly less than control group. The values of MPO, MDA and W/D were increasing as lung preservative period in vivo was prolonging. After 12h preservation, the value of MPO was significant changed while the value of MDA and W/D were changed significantly after 8h preservation. The pathological and super-micro structural changes were reversible till lung preservative period reached 8h.Conclusions: Method of modified self-confected LPD solution was simple and available. The effectiveness of lung preservation used with modified self-confected LPD solution was similar as LPD solution. Changes of data monitoring lung preservation started to be significant after 8h preserved with modified self-confected LPD solution and unreversible historical changes appeared after 12h preservation.
