Background:In recent years, combined heart and lung transplantation for end-stageheart and lung disease has been an effective treatment. But the long-termsurvival rate is still not satisfactory. Donor lung protection in the processof injury mechanism of lung injury and sensitive and effective marker hasnot been clear, donor lung preservation technology remains to beimproved.objective:This study was divided into two parts:1、Effect of Perfadex and Wisconsin Unviersity solution on the function of pulmonary artery endothelium2、Effect of Perfadex and HTK solution on the function of pulmonaryartery endotheliumMethods:Fifty-four porcine pulmonary rings in2mm long were randomly dividedinto three groups. Control group (n=18): Incubation in Krebs-Henseleit(KH) at4℃for4hours; group A (n=18) or B (n=18): Incubation inPerfadex or Wisconsin Unviersity solution (HTK solution for the secondpart of this study) at4℃for4hours. Endothelium-derivedhyperpolarizing factor (EDHF)-mediated relaxation (percentage of-7.5logM U46619precontraction) induced by bradykinin or calciumionophone A23187in the present of indomethacin (7μmol/L), L-NNA(300μmol/L) and oxyhemoglobin (20μmol/L)were measured in theorgan chambers.Results:1、Vasoconstriction induced by U46619is no significant differenceamong three groups(P>0.05).2、In the first part of this study, compared with control group, therelaxation induced by bradykinin or A23187was significantly decreasedin group A, while there is no significant difference in group B.3、In the second part of this study, the relaxation induced by bradykininwas no significant difference in group A(52.72±10.63%,P=0.14) or group B (54.24±9.33%,P=0.24), compared with control group(60.05±10.93%),. Meanwhile, the relaxation induced by A23187wasalso no significant difference between the control group and group A andB.Conclusions:1、Endothelium-derived hyperpolarizing factor (EDHF) plays animprotant role in endothelium-mediated relaxation of porcine pulmonaryartery.2、EDHF-mediated relaxation is imparied when the lung preserved withWisconsin Unviersity solution, wheras its function is not affected byPerfadex solution.3、EDHF-mediated relaxation showed similar performances when thelung preserved with HTK or Perfadex solution.
