Many researches have shown that Transforming Growth Factor β1 plays an important role in the propagating of cell and division , cell synthesizing and precipitation of ECM( Extracellular Matrix) , blood vessel of born and apoptosis of the cell.Developing the process inside, TGF - β1 in the tumor's occurrence has a double function; earlier period is taken placed in the tumor, and repress the growth of the tumor cell; then becomes the tumor incitement factor that grow along with the tumors development, i. e. the tumor cell can get away the repress from the TGF - β1 as to it s growth function. In recent years, along with peoples study continuously deep into as to it, discoveries of TGF - β1 can be used as the factor of many effects cells for having the activity of various biologies. In the tumor's occurrence, development, and invasion, now the domestic and international research seldom involve the TGF - β1 in the esophageal squamous cancer tissues in the inside 's expression and pathologic ratings, clinical periods, gradual depth of tumor, lymphoid transferring, distant - place transferring. This research comprises the proceeds of related study of this problem.Materials and MethodsEsophageal carcinoma tissues and surrounding esophageal tissues from 45 patients who received surgery at the First Affiliated Hospital of Medical University of China and Liaoning Tumor Hospital between September 2001 and March 2002 were frozen at - 80 . The patients ranged in the age from 36 to 73 years old ( averge age: 53. 2 years old ) ; male: 34; female: 11. They were all squamous cell carcinomas. 9, 22,11 And 3 of the cases were respectively classified as to TNM stage I , II , III and IV. The patients had no detectable metastasis in distant organs at the time of surgery. No patient had received chemotherapy or radiation therapy before surgery.1. Extraction of total RNA: Total RNA was extracted from tissue samples using a RNA extraction reagent, TRIzol reagent ( GIBCO-BRL, Int). The concentration of total RNA was adjusted to 1 ug/ul.2. Reverse Transcription (RT) ; 2p.l of total RNA were incubated in 12ul of reaction buffer (2ul MgCl2; 10 x RNA buffer 1. 25ul; Rnase Free dH2O 4. 5ul; dNTP 1ul; Rnase Inhibitor 0. 25ul; Reverse Transferase 0.5ul; Random 9 mers 0.5ul) that was purchased as an RNA PCR kit (AMV) Ver2. 1 (TaKaRa biology, China). The RT reaction was carried out in one cycle at 30℃ 10 min; 50℃ 30 min; 99℃ 5 min; 5℃ 5 min. 3. Polymerase Chain Reaction (PCR) : PCR primers (Sangon, China) for TGF β1 can detect TGF - β1 fragments according to the TGF - β1 gene structure. Aliquots of 6 ul first strand cDNA were mixed with 22 ul of PCR mixture. The PCR reaction was carried out in three steps as follows: 951 for 2 min (1 cycle) ; 94℃ for 1 min; 55℃ for 1 min; 12℃ for 1. 5 min (28 cy-cles) ; and 72℃ for 7 min( 1 cycle) .4. PCR fragments were analyzed by electrophoresis on 12% poly-acrylamide gels, and DNA was visualized by epithedium bromide staining.RESULTSAt 45 examples esophageal squamous cancer the specimen inside, each oesophagus cancer tissues the inside the TGF - B, the mRNA level all show the higher than normal buildup (P <0. 01) of cancer beside, but inside night period the esophagus cancer organize the inside the TGF - β1 the mRNA level show the higher than earlier period esophagus the cancer organize the inside the TGF - B, mRNA level (P <0.01) , but inside period and night period the esophagus cancer organize the inside the TGF - β1 the mRNA level more have no to show the difference ( P >0. 01) .TGF - β1 clinical and pathologic relation of mRNA level and e-sophagus cancer; the esophagus cancer that different buildup divide the degree organize the TGF - β1 of the insides the mRNA level have no to show the difference ( P > 0. 01). In the tumor cell the part is gradually depth inside, (T3 +T4) set TGF - β1 for esophagus cancer organizing the inside the mRNA level show the higher than ( T1 + T2) set.
