Selenium is one of the most important essential micronutrient elements in mammal, which mainly was shown to be incorporated as selenocysteine (Sec) in selenoprotein. Diabetes is one of the diseases which threaten human healthy in 21 century. The study of the relationship of selenium and diabetes has been paid close attention. It was reported that selenium can prevent and relieve diabetes and diabetic complications, but the mechanism is not clear. In this paper, the effect of selenium on the expression of glutathion peroxidase (GPx), thioredoxin reductase (TR), aldose reductase (AR), intercellular adhesion molecule-1 (ICAM-1) induced by high glucose or 3-morpholinosydnonimine (SIN-1) was investigated in Hepatoma cells (HepG2) and Human Umbilical Vein Endothelial cell (ECV-304) cells. The main results are as follows:1) The effect of glucose on the cell viability of HepG2 cells were investigated by 3-(4, 5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2H-tetrazolium bromide (MTT) assay, and the effect of glucose on the expression of GPx and TR in HepG2 was investigated by RT-PCR. The results showed that proliferation of HepG2 was inhibited by low dose of glucose (0~0.5 mM); it is supposed that the proliferation of HepG2 needs glucose, and the variation in the expression of GPx and TR induced by glucose is not significantly.2) The effect of selenium on the expression of AR, ICAM-1 in ECV-304 induced by high glucose or SIN-1 was investigated by real time RT-PCR. The result indicated that the expression of AR and ICAM-1 were increased by Se (0~100 nM) to maximum (50 nM), but the expression of AR and ICAM-1 induced by high glucose was inhibited by Se. When ECV-304 cell was treated with SIN-1, the expression of AR and ICAM-1 were increased (0~200μM), the optimization concentration is 100μM, which were different from the experiments reported. The difference maybe come from the dose which is used is lower than the reported. It is also observed that the expression of AR and ICAM-1 induced by SIN-1 was inhibited by Se.
