Objective To observe the effect of selenium,an antioxidant,on TNF-α induced expression of intercellular adhesion molecule-1 in rat vascular smooth muscle cells(VSMC) and to investigate the mechanism of atherosclerosis related to adhesion between monocytes and VSMC induced by TNF-α and of anti-atherosclerosis by selenium.Methods 6 years old's rat vascular smooth muscle cells were cultured in vitro,after incubated with TNF-α or TNF-α together with Se for 24h,Cell adhesion test wras employed,flow cytometric technique and RT-PCR were applied to measure the expression of ICAM-1 in protein level and mRNA level.Results TNF-a can increase the expression of ICAM-1 in vascular smooth muscle cells and Selenium of 0.5-2μM at 24h incubation could inhibit the adhesion of monocytes to VSMC and the expression of ICAM-1 in VSMC induced by TNF-a in a dose-dependent manner.Conclusions Selenium decrease monocytes adhesion to TNF-α-activated VSMC by inhibiting the expression of ICAM-1 in VSMC.This mechanism may contribute to the anti-atherosclerotic role of selenium.
