Study On Expression Of BRCA1 Associated Centrosomal Bacp/Nlp In Human Cervical Cancer

Background:Bacp/Nlp(BRCA1 Associated Centrosomal Protein/Ninein-like protein) locates at chromosome 20,is a novel protein associated centrosomal function,which was found through the Yeast two hybrid approach using the C terminal of BRCA1 by National Key Laboratory of Molecular Oncology,Cancer Institute,Chinese Academy of Medical Science.Preparation indicated that Bacp/Nlp gene was a potential oncogenic gene.Objective:The aim of this study is to investigate the levels of expression of Bacp/Nlp in tumor issue of cervical cancer by Immunohistochemical,Immunofluroescence, Western Blot and Reverse Transcription-Polymerase Chain Reaction(RT-PCR) technique,and these expression correlation with clinicopathologic parameters was analyzed.Methods:28 biopsy tissue with pathologically confirmed cervical cancer and 4 normal cervical tissue through total hysterectomy sample were eligible in Cancer Institute & Hospital Chinese Academy of Medical Science from Jan 2006 to Jan 2007.These expressions of Bacp/Nlp was investigated and detected by Immunohistochemical, Immunofluroescence,Western Blot and RT-PCR technique.And the correlation between Bacp/Nlp and clinical stage and pathological differentiation of cervical cancer was analyzed.Results:The positive expression of Bacp/Nlp was revealed in 42.9%and 25.0%of cervical cancer tissues but neither in normal cervical tissues by Immunohistochemical and Immunofluroescence,respectively,Western Blot and RT-PCR results showed that the expression of Bacp/Nlp in cervical cancer tissues was significantly higher than those in normal cervical tissues.The over-expression by Immunohistochemical showed no correlation with clinical stage and pathological differentiation.Conclusion:Bacp/Nlp is over expressed in cervical cancer at levels of protein and mRNA,Bacp/Nlp is not related to clinical stage and pathological differentiation.
