1.Experiment Study of the Influence on Growth and Apoptosis induced by Extract of Fungi of Huai'er in Human Pancreatic Cancer Cell Line Panc-1Objective: To investigate the growth inhibition and apoptosis induced by Extract of Fungi of Huai'er in human pancreatic cancer cell line Panc-1.Methods: Extract of fungi of Huai'er with different concentration gradient were cultured with human pancreatic cancer cell line Panc-1. Cell growth was assessed by MTT and FCM showed the change of cell apoptosis rate.Results: Extract of fungi of Huai'er induced a dose-dependent inhibition of human pancreatic cancer cell line Panc-1 in vitro. The maximum percentage of growth inhibition was17.7% at a concentration of 3mg/ml. The apoptotic ratio of human pancreatic cancer cell line Panc-1 increased parallelling with the increase of concentration of extract of fungi of Huai'er When the extract of fungi of Huai'er concentration was 3mg/ml, the apoptotic ratio reached (13.17±1.20)%, which was higher then that of group 6mg/ml(11.21±0.87)%, (P<0.05).Conclusions: Extract of fungi of Huai'er can significantly inhibit cell growth and induce apoptosis of human pancreatic cancer cell line Panc-1 in vitro, the optimal concentration is 3mg/ml.2. Clinical Research of Combination Treatment with Gemcitabine Chemotherapy and Huai'er Granula on Pancreatic CancerObjective: To study the clinical effect of Combination Treatment with gemcitabine(GEM) and Huai'er Granula on pancreatic cancer.Methods: In this prospective randomized controlled study, select 151 cases of pancreatic cancer(66 cases have undergone pancreectomy, 85 cases weren't resected)during Jul. 2004 to Sep. 2006, which included gemcitabine chemotherapy [33 cases (pancreectomy), group a + 42 cases (weren't resected), group A], gemcitabine+Huai'er Granula [33 cases (pancreectomy), group b + 43 cases (weren't resected), group B] and plus blank group [17 cases (pancreectomy), group c + 23 cases (weren't resected), group C], compared clinic effect of each group.Results: The blood side-effect of group GEM and GEM+Huai'er were 76.32% and 61.84% after treatment, the difference was significance (P<0.05). After chemotherapy, the group GEM+ Huai'er of the levels of IgG and IgM, the percentage of Nk cells and CD3+, rate of CD4+/CD8+ were higher than group GEM, the difference were significance (P<0.05). The rates of clinical benefit response of group GEM , GEM+ Huai'er were higher than the blank group(P<0.05), on the 3rd month, the difference of group GEM+ Huai'er compared GEM was no significance (P>0.05); on the 6th month, the difference of group GEM+ Huai'er were higher than group GEM (P<0.05). The rates of objective tumor response of group A, B were higher than group C(P<0.05), but the difference between group B and A was no significance (P>0.05). The recurrence and metaptosis rate of group a, b and c have no significant difference(P>0.05). there were no significant difference between GEM chemotherapy and GEM+Huai'er combined therapy in 1 year survival rate (P>0.05).Conclusions: Treatment of pancreatic cancer with gemcitabine chemotherapy combined Huai'er Granula improved the rates of objective tumor response , recurrence and metaptosis and survival rates of 1 year didn't have statistical evidence, but it improved side-effect of chemotherapy, survival quality and immune function.
