Objective:To compare the inflammatory responses induced by poly(ether-ether-ketone)(PEEK),traditional ultrahigh molecular weight polyethylene(UHMWPE,referred to as PE)particles and cross-linked ultra-high molecular weight polyethylene(x PE)particles in vitro and to provide further experimental basis for the selection of new marerials for clinlical joint injury repair.Methods:Using the osteoclast precursor cell-mouse peritoneal macrophage cell line(RAW264.7)as the experiment object,find the appropriate ratio of cell and particle.The effects of three different particles of PE(particle size,5.5 ?m),x PE(particle size,0.7 ?m)and PEEK(particle size,0.7 ?m)on cell proliferation were measured by Cell Counting Kit-8(CCK-8).Cytotoxicity LDH Assay Kit-WST(LDH)was used to detect the toxicity of particles to cells.After treatment with PE,xPE and PEEK particles,the protein levels of TNF-?,IL-6 and IL-1? were determined by the ELISA,and the gene expression levels of TNF-?,IL-6 and IL-1? were examined by qPCR.To find the preliminary mechanism,the expression of nuclear factor ?B receptor activator of nuclear factor ?B(RANK)gene in RAW264.7 cells was also detected by qPCR technique.Results:CCK-8 and LDH results showed that the suitable ratio of cells to particles was 1:20.The particles of PE,xPE and PEEK resulted in a certain growth inhibition on RAW264.7 cells.CCK-8 showed that particles had significant growth inhibitory effect after treatment at different time,but LDH showed that particles had no obvious toxic effects.At the same time,from the protein level by ELISA and the transcription level by qPCR,the levels of TNF-?,IL-6 and IL-1? in PE group and xPE group were significantly higher than those in the control group(except the IL-1? protein level),but the expression level of PEEK treatment group was significantly lower than those of other treatment groups.The expression of RANK gene was detected by qPCR,and the PEEK group expression level was also lower than that of PE or x PE group.Conclusion:The inflammatory response caused by PEEK material wear particles,compared to PE and xPE two materials,was significantly lower,and it may be better used in clinical application of joint prosthesis.
