Objective To investigate the anti-tumor effect of tanshinoneⅡA on the human gastric cancer cell line SGC-7901 and its machanism.Methods The human gastric cancer cell line SGC-7901 was treated with tanshinoneⅡA at various concentrations . The effect of cell growth inhibition was assayed by MTT method.Cell cycle,apoptosis and the expressions of p53,bcl-2 were assessed by flow cytometry .Results TanshinoneⅡA could inhibit the growth of the human gastric cancer cell line SGC-7901. The inhibitory effect was dose-and-time dependent. There was an apoptosis peak detected by flow cytometry,cell cycle was arrested in G0/G1 phase. After treatment with tanshinoneⅡA (0.5,1.0,2.0,4.0μg/ml ) for 72h,the apoptotic rates were 26.97±2.68%,31.52±4.36%,40.21±3.56% and 42.18±3.84%, respecteively (P<0.01), bcl-2 gene expression down-regulated and p53 gene expcession up-regulated were observed.Conclusion TanshinoneⅡA could inhibit the growth of human gastric cancer cell line SGC-7901 in a dose-and-time dependent manner. Its possible mechanism might relate to arrest cells at G0/G1 phase , induce cell apoptosis ,upregulate p53 and downregulate bcl-2 gene expressions.
