Objective To observe the effects of SHENXUENING JIANJI on clinical symptoms,24 hours urine protein quantity, the levels RBC, kidney morphology, renal function of model rats,and the expression of connective tissue growth factor(CTGF) in kidney. To observe the anti-renal fibrosis effect of Chinese traditional medicine SHENXUENING JIANJI.To explore the mechanism of tradictional Chinese drugs treatment on IgAN. To find the new target spot on renal fibrosis.To explore the mechanism of action of Chinese traditional medicine on renal fibrosis.Methods The model of IgAN was established by feeding the Wistar famale rats Borine Serum Albumin (BSA) and injecting BSA and Staphylococcal Entertoxin B(SEB) in their caudal veins.We observed the levels RBC and protein of urine, renal function and pathological change of experimental rats. The expression levels of CTGF in kidney were determined by immune chemical staining methods.Then we investigated the expression of CTGF using image analysis.Results①After treatment, the clinical symptoms of two treatment groups were improved compared with model group ,SHENXUENING JIANJI group was the better。②the levels RBC,24h protein urinary quantification,the levels of Scr and BUN in model group were higher than those in control group (P<0.01);After treatment, every indexe of SHENXUENING JIANJI group was improved. but there was no significant difference compared with 24h protein urinary quantification in SHENXUENING JIANJI group and Valsartan group(p>0. 05), there was significant difference compared with the levels RBC in SHENXUENING JIANJI group and Valsartan group(P<0.01),there was no significant difference compared with the levels of Scr and BUN in SHENXUENING JIANJI group and Valsartan group(p>0. 05).③Renal glomerulus diffuse hyperplasy , The extracellular matrix and mesangial cell had hyperplasy of moderately and seriously widespread mesangial matrix.The most capsular of basement membrane of renal glomerulus irregularity,a small quantity renal tubule atrophy, interstitial inflcell infiltrated and focal fibrosis,and renal tubuleke grana vacuolar degeneration.Pathological changes display- ed the class ofⅢorⅣ.Kidney irnmunofluorescence shows model group has severe, widespread IgA deposition in the mesangial area.while two treatment groups are improved compared with model group.Pathological changes displayed the class ofⅠorⅡ.Kidney irnmunofluorescence shows model group has positive or moderate, widespread IgA deposition in the mesangial area.④The expression of CTGF in kidney of model group was significant higher compared with control Group(P<0.01), and tubulointerstitial was higher compared with renal glomerulus.After treatment,the expression of CTGF in kidney of SHENXUENING JIANJI group and Valsartan group were improved, there are significant difference when compared with the model group (P<0.01),but there was no significant difference compared with control group(p>0.05).Conclusions SHENXUENING JIANJI can not only improve signifi- cantly IgAN clinical symptoms and reduce the levels RBC and protein urinary quantification and protect renal function but also improve renal pathology, even inhibit the expression of CTGF in kidney. And then delay renal fibrosis (renal interstitial fibrosis).
