Objective: To investigate the expression of sphingosine kinase-1(SphK1),sphingosine 1-phosphate receptor(S1PR)and alpha smooth muscle actin(?-SMA)in the kidney tissues of IgA nephropathy(IgAN)patients.Search for the function of SphK1 pathway in the occurrence and development of IgAN,to provide experimental basis for clinical treatment of IgAN.Methods: 18 kidney tissues of IgAN patients were selected as experimental group,and 5 kidney tissues removed from the normal tissues of the Renal tumor patients were control group.Collected the clinical data of the patients in both two groups,including 24-hour urine protein,serum creatinine,urea nitrogen and so on.Immunohistochemical method was used to detect the expression of SphK1,S1PR1,S1PR2 and ?-SMA in renal tissues.Image-Pro Plus(image analysis software)was applied to analyze the OD value of these images.Investigate the relationship of SphK1 with ?-SMA and clinical datum.All data were analyzed by spass18.0,the measurement data are expressed as mean(±)standard deviation((?)±s),the experimental group and the control group were compared by t test of independent samples,correlation analysis was tested by Pearson,p<0.05 showed that the difference was statistically significant.Results: 1.The positive levels of SphK1 ? S1PR2 ? ?-SMA and S1PR1/S1PR2 in renal tissues of patients with IgAN were higher than those in control group,there are significant difference between them(P<0.05);Thepositive level of S1PR1 is no difference between IgAN and control group(P>0.05).2.The level of SphK1 was positively correlated with ?-SMA ?24-hour protein and the serum creatinine(p<0.01 or p<0.05).There was no linear correlation between SphK1 and blood urea nitrogen(p>0.05).Conclusion: 1.SphK1 pathway may be involved in the pathogenesis of IgAN;2.The role SphK1 signal channel plays in IgAN may related with the mesangial cell phenotype change caused by ?-SMA;3.There may be S1PR1/S1PR2 differential transduction during the binding of S1 P to it's receptor subtype;4.The expression level of Sph K1 in renal tissue can reflect the degree of renal damage to some extent.
