Clinical And Experimental Research On The Wei Tiao San Hao Decoction Treating Advancing Colorectal Cancer

Purpose:To observe the clinical effect of Wei Tiao San Hao decoction on the physical strength,clinical symptoms,new future curative effect,serum tumor sign, chemotherapeutic toxicity in curing advancing colorectal cancer and to research the effect of Wei Tiao San Hao decoction on growth suppression and apotosis of HT-29 and Lovo colorectal cells by serum pharmacology method.Methods:Sixty cases of advancing colorectal cancer were chosen and divided into two groups,trail group and control group.Thirty cases of trail group were treated with Wei Tiao San Hao decoction and chemotherapy with OLF scheme,another thirty cases of control group were treated with the same scheme only.Compare the two groups in physical strength,clinical symptom amelioration,the level of serum CEA and CA19-9,new future curative effect,and degree of side effects caused by chemotherapy,and count out the difference in the upper aspects between the two groups.We used Sulforhodamine B sodium salt(SRB) to observe the growth suppression of HT-29 and Lovo colorectal cells.We researched the cell apoptosis through morphologic observation with phase contrast microscope,result of Hoechst 33258 dyeing,and examination of cell apoptosis used Annexin V-FITC reagent.Results:The trail group displayed more effective in enhancing the physical strength, releasing the cilnical symptoms,alleviating the side effects caused by chemotherapy than the contral group.In new future curative effect,the effective rates was similar to the control group,the disease control rates was higher than the cohtral group,the difference between them was distinct.The two groups had the similar effect on the level of serum CEA and CA19-9.The experiment of cell growth suppression suggested that the WD-3 rabbit serum displayed more effective in inhibiting cell growth than the normal rabbit serum in the concentration range from 10%to 40%,the difference between them was distinct,but they had similar effect when the concentration exceeded 40%.The morphologic observation with phase contrast microscope,result of Hoechst 33258 dyeing and examination of cell apoptosis used Annexin V-FITC reagent suggested that the WD-3 rabbit serum displayed more effective in cell apoptosis than the normal rabbit serum. Conclusion:In the treatment of advancing colorectal cancer,Wei Tiao San Hao decoction proved its function in enhancing the physical strength,releasing the cilnical symptoms,stabling the tumor and improving the disease control rates,alleviating the side effects caused by chemotherapy.Wei Tiao San Hao decoction could induce apoptosis of HT-29 and Lovo colorectal cells and inhibit cell growth.
