Objective: 1.Discussing the correlation between Traditional Chinese Medicine Syndrome Types of Chronic Hepatitis B (CHB) and the change of liver histopathology. 2. An orientation study of relations between DNA ploidy on liver cell and syndrome type of Traditional Chinese Medicine.Approaches: 1. Select a total of 413 cases needle biopsy of liver with Chronic Hepatitis B in the liver centre of Xiamen Tranditional Chinese Medicine Hospital,collect up and adopt clinical syndrome types of Traditional Chinese Medicine (T.C.M) in the patients with CHB, observe the liver tissue slices which are collected up and selected, record the pathological grade and stage, meanwhile conduct the semi-quantitative scoring according to the new scoring system which is improved by Wan Tailing on the basis of Knodell's Histological Activity Index system and Chevalier's semi-quantitative scoring system. Analysis the correlations among the syndrome types of T.C.M, the histopathological grade and stage and the histological semi-quantitative scoring. 2.Random select 181 cases from the cases above-mentioned: collect up and select clinical syndrome types of T.C.M, and conduct Feulgen (FG) staining on the tissue slices that been selected, use image analysis system to conduct the content test of DNA, ploidy analysis and cell cycle test in liver cell, analysis the correlations among content of DNA in liver cell, DNA Index in liver cell and Proliferation index in liver cell.Results: 1. The correlation between syndrome types of T.C.M and inflammation of hepatic tissue also the correlation between syndrome types of T.C.M and the degree of fibrosis: 1.1 The syndrome types of T.C.M: the three common types: there are 204 cases belong to liver stagnation and spleen deficiency type (47.11%), there are 173 cases belong to damp-heat mid-retention type (39.95%), there are 56 cases belong to the type of blood stasis hindering in the channels (12.93%). 1.2 According to histopathology, there are 286cases belong to low-grade chronic Hepatitis (66.05%), there are 108cases belong to moderate chronic hepatitis(24.94%), there are 39cases belong to serious chronic hepatitis(9.01%).1.3 The syndrome types of T.C.M have obvious correlations with pathological inflammation grading, fibrosis staging, inflammation and semi-quantitative scoring system. Among the three types above, the degree of liver stagnation and spleen deficiency type is the lowest, also the degree of blood stasis hindering in the channels is the highest.3. The analysis of the correlation between syndrome types of T.C.M and content of DNA in liver cell, also between syndrome types of T.C.M and ploidy. 2.1 There are three common syndrome types of T.C.M: there are 73 cases belong to liver stagnation and spleen deficiency type (40.33%), there are 68 cases belong to damp-heat mid-retention type (37.57%), there are 40 cases belong to the type of blood stasis hindering in the channels (22.1%). 2.2 In the aspects of the research index coming from the DNA content in liver cell, ploidy distribution and cell cycle. By means of statistics, comparing random two from the DNA content in liver cell, ploidy distribution and cell cycle, there is significant difference among all of them except aneuploid cell percentage, the DNA content, DNA Index, Proliferation index of the three syndrome types of T.C.M above is all increasing orderly. (liver stagnation and spleen deficiency type < damp-heat mid-retention type < the type of blood stasis hindering in the channels).Conclusions: There is significant correlation between the common syndrome types of T.C.M and inflammation of hepatic tissue, also between those types and the degree of fibrosis, also between the types and DNA content in liver cell.
