Background:More clinical common diseases can lead spinal cord injury,as Scoliosis, lntraspinal Occupying Lesion,Anterior Spinalartery Syndrome,Posterior Spinal Artery,all of could engender primary disfunction of spinal cord.The second injury of spinal cord after reperfusion is always a difficult problem of medical.The lately reports have show that incidence of spinal cord ischemia eperfusion injury is 3%~18%assumably.The mechanism of Spinal Cord lschemia is still unknown.MP as the top choice medicine of healing or protective effects of Spinal Cord Ischemia,however,high dose MP was treated can bring more complications,include infection,bedsore,Gastrointestinal bleeding,Deep venous thrombosis.The former experiment shows that Honghua injection play alleviative roles in spinal cord ischemia and reperfusion.Objective:To observe the effects of different doses of honghua injection on spinal cord ischemia and reperfusion injury at neurological function score,histopathological changes, immunohistochemical changes and neurological cell ultrastructure on TEM.,and to investigate the better dose of Honghua injection in Spinal Cord Ischemia and Reperfusion Injury,with the purpose of providing experimental testimony to clinical treatment.Methods:New Zealand white rabbits(n=90) were randomly allocated to 3 groups:Group A (with low dose,n=30),Group B(with middle dose,n=30),Group C(with high dose, n=30).Spinal ischemia reperfusion injury was made by cross-clamping infrarenal aortic for 35rain according to Zivin's method.Group A injected 2.5ml/Kg Honghua injection through vein before aortic cross-clamping;Group B receives 5.0 ml/Kg and Group C treated with 10.0 ml/Kg as the same method.At 4h,24h,48h,72h,7d after reperfusion,6 rabbits of each group radomly were oberserved to investigate the neurological status,they were killed to remove the L3-5 spinal cord quickly,Cell apoptosis TUNEL labeling were studied and the expression of caspase-3 was accessed by immunohistochemical staining,neurological cell ultrastructure was oberserve on TEM 7d after reperfusion.Result:Three groups were observed post cells of TUNEL labeling and Caspase-3 expressed, they were present with a maximum presence at 24h after injury and decreased from 48h.A positive correlation between expression of Caspase-3 and apoptosis was found.The number of positive cells stained TUNEL labeling and caspase-3 expressed in the middle-dose group was smaller than other group at every time point,the average neurological function score was significantly higher than that in low-dose group and high-dose group at all phase points,but histopathology changes was no significant.Conclusion:The middle-dose honghua injection has higher effectiveness than other does in Spinal Cord Ischemia and Reperfusion Injury in rabbits,which can as dose Reference to clinical treatment.
