Background:Mitochondrial DNA(mtDNA) is the only germ out of nuclear DNA;It easily becomes attacked target by carcinogen and ROS,because of lacking histone protecting and effectual repairing system.The antioxidative system is the main system of eliminating ROS,and is related with the Mitochondrial.It is going a step further to study,that mtDNA is related with the antioxidative system and what signal conduction way is.Objective:1) To examinate the copy numbers of mtDNA in blood of the breast carcinoma;2) To examinate the change of the GSH-Px,SOD and MDA of the antioxidative system in blood of the breast carcinoma;3)To detect the relationship of the copy numbers of mtDNA and the GSH-Px,SOD and MDA in blood of the breast carcinoma.Methods:1) The HV1 and HV2 of the mitochondrial D-loop region from 25 cases breast carcinoma blood and 18 cases health control group were amplified by PCR; meantimeβ-actin was served as a quantitative standard marker,followed by polyacrylamide gel electrophoresis(PAGE) and silver staining,in which the difference of mtDNA copy number was compared between breast carcinoma and control group blood;2) The GSH-Px,SOD and MDA rom 25 cases breast carcinoma blood and 18 cases health control group were amplified by the Biochemistry Examination,in which the difference of them were compared between breast carcinoma and control group blood.Results:1) There were lower of the copy numbers of mtDNA in the breast carcinoma blood than the control group blood(P<0.05);It was the positive related of the HV1 and HV2(P<0.01);And it was higherⅠstage thanⅡstage thanⅢstage of TNM;2) There were lower of the GSH-Px and SOD in the breast carcinoma blood than the control group blood(P<0.01),and higher of the MDA in the breast carcinoma blood than the control group blood(P<0.01);The GSH-Px and SOD was higherⅠstage thanⅡstage thanⅢstage of TNM(P<0.05),and it was not related with the Lymph node transfer(P>0.05);The MDA was higher in the positive Lymph node transfer than the negitive Lymph node transfer(P<0.05),it was lowerⅠstage thanⅡstage thanⅢstage of TNM(P<0.05).Conclusions:1) There were lower of the copy numbers of mtDNA in the breast carcinoma blood than the control group blood,which may be a new tumor marker.2) The antioxidative system in blood of the breast carcinoma was damaged,and were related with the Lymph node transfer and TNM.3) the copy numbers of mtDNA was related with the antioxidative system,there may be some ways between them.
