Background:The mitochondrial DNA (mitochodrial, mtDNA) as the only outside nuclear genetic material has been found with a variety of diseases and a variety of tumor occurrence and development. mtDNA ATPase6mitochondrial genome encoding complex V largest subunit gene, which together with the mtDNA ATPase8encoding ATP synthase FO part. As the mitochondrial genome encoding the FO of the guiding role of the energy produced there, so the research ATPase6mutations in breast cancer tissues and expression of the understanding of breast cancer, tumor cell development and modes of supply of energy will certainly help.Objective:1) Detection of invasive breast cancer gene mutation mtDNA ATPase6to find specific mutations;2) Testing mtDNA ATPase6in invasive breast cancer in the transcription level, to explore its in cancer development particularly in the energy supply role.Methods:The polymerase chain reaction (PCR) with gene sequencing method,20cases of invasive breast cancer gene sequencing for mtDNA ATPase6; Reverse Transcription-Polymerase Chain Reaction (RT-PCR), detection of invasive breast cancer20cases,20cases of cancer tissue mRNA expression of mtDNA ATPase6, and the nuclear genome of the β-actin as a quantitative standard material, comparing the two genes in cancer tissue and adjacent tissues of the differences.Results:20cases of breast cancer genes were found in mtDNA ATPase611base sequence variants, of which six genetic polymorphisms, five base variations are mutation frequency of5or more variable sites53, namely:C8673T, A8729G, T8955C;20subjects were in the mtDNA ATPase6the level of transcription in cancer tissue as ATPase6/p-actin±S value of0.732±0.137, as in cancer tissue ATPase6/β-actin±S value of0.344±0.125, comparison of the two are significantly different p=0.005<0.01. With clinicopathological parameters and found that the transcription level ATPase6TNM-I period of less than TNM-Ⅲ period p<0.005, statistically significant.Conclusion:1. invasive breast cancer mtDNA ATPase6the presence of high mutation C8673T, A8729G, T8955C, the incidence of breast cancer and may play an important role in development;2. invasive breast cancer in mtDNA ATPase6transcription levels, the tumor cells a high demand for energy performance;3. from the energy supply point of view, mtDNA ATPase6transcription levels of invasive breast cancer there is an alternative way to provide the basis for energy supply.
