Studying The Relationship Between The Character Of Diffusion-weighted Imaging And The Expression Of PCNA And MMP-9 In Gliomas

Abstract/Summary:
Objective: To study the value of diffusion-weighted imaging (DWI) in identifying different grade brain gliomas. To evaluate the expression of proliferating cell nuclear antigen (PCNA) and matrix metalloproteinase-9 (MMP-9) in different grade brain gliomas and analysis the relationship between relative apparent diffusion coefficient (rADC) of gliomas and the expression degree of PCNA and MMP-9 in gliomas.Methods: 30 patients with brain gliomas underwent conventional MR imaging and diffusion-weighted imaging (with b values of 0 s/mm2 and 1000 s/mm2) before operation. All of these cases were pathologically confirmed, there were 10 low-grade gliomas (grade 1 and 2) and 20 high-grade gliomas (grade 3 and 4). ADC maps were calculated by the workstation. ADC values of solid tumors components, adjacent edema region, far peritumoral edema region and opposite normal white matters were measured and the rADC values were calculated. And the expression of PCNA and MMP-9 were detected in these 30 cases by immunohistochemical (SP) staining.Results: (1) rADC values of low-grade gliomas (1.77±0.33) was higher than that of high grade gliomas (1.03±0.28) (p<0.05). (2) rADC values of adjacent peritumoral edema region (1.98±0.31) was lower than far peritumoral edema region (2.73±0.24) (p<0.05). (3) The expression degree of PCNA and MMP-9 were significantly higher in high-grade gliomas than that in low-grade gliomas (p<0.005). (4) The rADC values of solid tumors components was correlated with the negative expression of PCNA (p< 0.001). The rADC values of adjacent and far peritumoral edema region were correlated with the positive expression of MMP-9 (p<0.001).Conclusion: (1) The rADC values of solid tumors components can provide useful information in identifying the grade of gliomas. (2) The rADC values of adjacent peritumoral edema region was lower than far peritumoral. It suggests that adjacent peritumoral edema region is more obviously to be invased. (3) The rADC values of solid tumors components can show the proliferating station of gliomas. (4) The rADC values of peritumoral edema region can provide useful information of aggressiveness of gliomas.
