Objective:Establish a stable, good SD rats experimental model with C6braingliomas. To confirm the performance of self-developed small sizedmedical imaging spectrometer depending on the imaging spectrum andimaging data of brain gliomas in SD rats through online real timescanning this animal model with the imaging spectrometer. This researchcan help lay the foundation for studying about medical imaging spectrumtechnology.Method:(1)Preparation of experimental animal models:①Laboratoryanimals:37healthy clean SD rats, pure lines, weighing200~250g,17malead and20female.②Cultivation of rat gliomas cell lines C6cells.③Animal model:10%chloral hydrate (0.35mg/100kg) to the SD ratperitoneal administration of anesthesia. Fix the head with stereotaxicapparatus after anesthesia works. Both auricular needles go deep intoexternal auditory canal and fix symmetrically, cut off the hair on top ofhead, unfold drapes after sterilization with iodine tincture and alcohol.Incise the scalp about1.5cm longitudinally along with middle sagittal line,separate the exposured parietal bone fully, drill hole with dentist’smicro-drill at coronal suture rear1mm,3mm right side of centre line with guide of stereotaxic apparatus without harming endocranium. Expand thehole to a bone window. Get10ul C6cell suspension with concentration of1×106/μL, fix it over stereotaxic apparatus, vertically insert needle toendocranium6mm through the centre of done window slowly and1mmback(5mm to endocranium) by10ul microsyringe, Finish injecting in30s,retain the needle1min, exit the syringe in10s to make cells precipitatedfully. Close the bone window with sterile bone wax, wash surgery fieldwith normal saline, sew up the incision with line4, disinfect the skin.(2)Changchun Institute of Optics, Fine Mechanics and Physics, ChineseAcademy of Sciences design and develop one small sized medicalimaging on the basis of combination of spectroscopic techniques ofprism-grating-prism, which can be integrated with operating microscope.(3)Collect imaging spectrum data of brain glioma in SD rats, we canjudge where and how large the cerebral tumor is through the analysis onthe imaging spectrum data.Results:①37SD rats are divided into three batches, only31SD rats gettumor as it is affected by multi factors in constructive process, tumor rate83%.②Changchun Institute of Optics, Fine Mechanics and Physics,Chinese Academy of Sciences design and develop one small sizedmedical imaging on the basis of combination of spectroscopic techniquesof prism-grating-prism, which can be integrated with operatingmicroscope.③Analyze the imaging spectrum data collected from onlinereal time scanning to animal experimental model with the imaging spectrometer, which can judge the tumor tissue boundaries exactly andshow operator with pictures.Conclusion:①Prepare animal experimental model of SD rat with brain gliomasuccessfully.②Changchun Institute of Optics, Fine Mechanics andPhysics, Chinese Academy of Sciences design and develop one smallsized medical imaging on the basis of combination of spectroscopictechniques of prism-grating-prism, which can be integrated withoperating microscope.③Analyze the imaging spectrum data collectedfrom online real time scanning to animal experimental model with theimaging spectrometer, which can judge the tumor tissue boundariesexactly and show operator with pictures. This instrument is noninvasive,no radiation, high accuracy positioning online and real-time, which canhelp surgery of accurate tumor resection effectively. This experiment willpromote the application of imaging spectroscopy technology further inthe field of clinical diagnosis in our country and lay a foundation for theresearch of medical imaging spectrum technology.
