Objective⒈To observe the immunotherapeutic effects of hα-syn DNA vaccine in chronic parkinson's mice induced by little dose of 1-methyl-4-phenyl- 1,2,3,6-tetrahydropyridine(MPTP) injection.⒉To observe the immune inflammatory reaction of hα-syn DNA vaccine in chronic parkinson's mice induced by little dose of MPTP injection.MethodsThe constructed hα-syn DNA vaccine was injected intramuscularly to chronic parkinson's mice as the test group, the other two groups of mice were set up as control including vacant vector plasmids and PBS injected. The expression changes ofα-syn, tyrosine hydroxylase(TH ),CD11b and TNF-αin Substantia nigra was visualized by immunohistochemistry. The immmunotherapeutic effects in chronic Parkinson's mice were assessed by the behavioral changes in mice and the expression changes ofα-syn and TH in substantia nigra. The immune inflammatory reaction in chronic Parkinson's mice were assessed by the expression changes of CD11b and TNF-αin substantia nigra.Results⒈Behavior observation results: compared with the control groups, the climbing pole time of the test group was obviously decreasing (p<0.01), and those parkinsonism behaviors of the test group improved markedly.⒉Morphologic examination results: after HE dyeing, compared with the control groups, the number of dopaminergic neurons and microglia cell of the test group increased obviously.⒊SP immunohistochemistry results ofα-syn and TH: Compared with the control groups, masculine dyeing ofα-syn of the test group decreased, the masculine expression ofα-syn of the test group decreased, displayed marked difference(p<0.01).The TH expression of the test group increased, the counts of TH positive cell of the test group increased obviously, CPC werel markedly different with the control groups(p<0.01).4.SP immunohistochemistry results of CD11b and TNF-α: Compared with the control groups, masculine dyeing of CD11b and TNF-αof the test group increased, The CD11b and TNF-αmasculine expression of the test group increased, displayed marked difference(p<0.01).Conclusions1.hα-syn DNA vaccine have a better therapeutic effect in chronic Parkinson's mice. Immunization with hα-syn DNA vaccine yielded a 30 % ofα-syn expression reduction rate and a 31%-39% of dopamine neuron counts increasing rate,showing a significant different compared with those of control groups(P<0.01).2.hα-syn DNA vaccine have some immune inflammatory reaction in chronic Parkinson's mice. Immunization with hα-syn DNA vaccine yielded TNF-αexpression increasing and activated microglia cell increasing, showing a significant different compared with those of control groups.
