Objective:1.To compare the concentration of PM2.5in the air collected from the polluted area by Ni and the control area that have the similar background of culture and environment except Ni.2.To survey the effects on rat's cardiovascular system caused by collected PM2.5from Ni polluted area and the control area.Methods:1.The PM2.5from the Ni polluted area and the control area were collected day by day.2.40 male rats were randomly divided into four groups and respectively exposed to the PM2.5from the Ni polluted area and the control area,nickel sulfate and saline by intra-tracheal instillation.Rats were killed 24h after treatment.The levels of IL-6,CRP,sICAM-1,sVCAM-1 in blood serum and MCP-1 in tissue homogenete of heart were examined.Result:1.The result of PM2.5in the Ni polluted area and the control area:①The concentration of PM2.5in the Ni polluted area and the control area were 91.00±48.51 and 67.20±18.48(mg/m3)in summer and 0.057±0.015和0.134±0.021 (mg/m3)in winter,and there was no significant difference between Ni polluted area and the control area.②The concentration of Nickel in the collected PM2.5from the Ni polluted area and the control area were respectively 0.195μg/m3 and 0.004μg/m3,and the concentration of Nickel in Ni polluted area was almost 50 times than the control area.2.The results of acute toxic effects caused by PM2.5to cardiovascular system of rats:①The contents of IL-6 in blood serum of rats were elevated after being exposed and there was dose-effects relationships,and the elevated sequence was Ni polluted area>control area>nickel sulfate;②The contents of CRP in blood serum of rats were elevated after being exposed and there was dose-effect relationships,and the elevated sequence was the same as the change of IL-6;③The contents of MCP-1 in heart tissue homogenate of rats exposed to PM2.5from the Ni polluted area was significantly higher than that from control area and nickel sulfate,but there was no significant difference between control and nickel sulfate;④The contents of sVCAM-1 in blood serum of rats exposed to PM2.5from the Ni polluted area was significantly higher than that from control area and nickel sulfate in middle dose,but there was no significant difference in other doses;there was no significant difference in the contents of sICAM-1 in blood serum of rats among all groups.Conelusion:1.There was no significant difference in concentration of PM2.5between Ni polluted area and the control area.But the concentration of Nickel in the collected PM2.5from the Ni polluted area was almost 50 times than the control area;2.Inflammation reaction was caused by being exposed to PM2.5and the Inflammated biomarkers of cardiovascular system were elevate,and the sequence was Ni polluted area>control area>nickel sulfate;3.The results indicated there was potential hazard to cardiovascular system by Nickel in the PM2.5.
