Laryngeal cancer is a common head and neck malignant tumor. The traditional treatment is surgical resection, and giving auxiliary radiotherapy according to patients condition at the same time. Chemotherapy or chemotherapy combined radiotherapy could improve the overall survival and organ retention for recurrence of laryngeal cancer patients. Celecoxib has been approved by FDA to be used in the treatment of pain, and reduce the number of adenomatous colorectal polyps in patients with familial adenomatous polyposis (FAP). Recent studies in vitro suggested that celecoxib, a selective COX-2inhibitor, can inhibit tumor growth and decrease the activity of COX-2. In short, this study found celecoxib is a kind of promising laryngeal cancer chemotherapy drugs. The present study was designed to investigate the possible role of celecoxib against laryngeal cancer.Objective This study was aimed at investigating the cellular level to show the ability of celecoxib to inhibit larynx carcinoma cell lines growth, by inducing cell cycle arrest and apoptosis. What’s more, we attempted to demonstrate the mechanisms underlying the chemopreventive effect of celecoxib against laryngeal cancer.Methods MTT was used to observe the proliferation of Hep-2cells treated with various doses celecoxib at different hours. Cell apoptosis was determined by observing the ultrastructure under electron microscope, flow cytometry,and fluorescent microscopy with Hoechst33258staining. Changes in mitochondrial membrane potential were detected with JC-1staining. Using flow cytometry examined the cell cycle distribution. Protein expression was examined by western blot.Results Celecoxib inhibited the proliferation and induced Go/Gi arrest, apoptosis,and loss of mitochondrial membrane potential in Hep-2cells. Western blot results showed celecoxib can down-regulate the levels of expression of COX-2and Bcl-2protein, up-regulate Bax, p27KIP1, P16INK4a protein’s expression, activate caspase-3and PARP, and transport AIF from mitochondria to nucleus.Conclusion Results from the present study indicate that celecoxib exerts its anticancer effect through inducing apoptosis and cell cycle arrest in Hep-2cells and may be a kind of promising laryngeal cancer chemotherapy drugs.
