Objective To investigate the role of dendritic cells in the pathogenesis of Henoch- Schonlein Purpura in children.Methods Thomas'method was adopted.The adherent precursors of DC were isolated from peripheral blood of Henoch-Schonlein Purpura's children and healthy controls.The adherent cells were induced with granulocyte-macrophage colony-stimulating factor(GM-CSF),interleukin-4(IL-4)and tumor necrosis factor-a (TNF-a)to DC in vitio.The expression of the surface molecules CD80,CD86 on the DC was examined by fluorescent activated cells sorter(FACS),and the ability to secret IL-6,IL-12 and IL-10。And their potentials to stimulate the proliferative reaction of DC inductive self T-Iymphocyte were investigatethed.Results The results showed that the Henoch- Schonlein Purpura's children had remarkably increased the ability to secret IL-6 compared with normal control subjects (p<0.01).,While the ability to secret IL-12 remarkably decreased compared with normal control subjects(p<0.01),the Henoch-Schonlein Purpura's children had remarkably decreased the ability to secret IL-10 compared with normal control subjects(p<0.01);meanwhile,the co-stimulating factor CD86 expressed by DCs remarkably increased in the Henoch-Schonlein Purpura 'children compared with normal control subjects(p<0.01),and self T-Iymphocyte proliferative reaction induced by DC was remarkably increased compared with normal control subjects (p<0.01).Conclusion Dc possibly plays a vital role in the immunological mechanism of Henoch- Schonlein Purpura by means of inducing the differentiation of Th1/Th2,the possible mechanism may involve interleukins(especially IL-6,IL-10 and IL-12)secreted by DCs.At the same time,in Henoch-Schonlein Purpura's children,self T- Iymphocyte proliferative reaction induced by DC was remarkably increased compared with normal control subjects(p<0.01),also can induc the differentiation of Th1/Th2,and have important immunomodulator function.
