Pre-eclampsia(PE)is the specific pregnant disease that results in significant perinatal morbidity and mortality.The disease is manifest in the third trimester of pregnancy with a wide variety of maternal symptoms,of which hypertension,proteinuria,and edema are late manifestations of a multi-factorial,multi-systemic disorder initiated in early pregnancy.The pathogenesis of PE is very complicated for many elements play roles in the disease.More and more evidences show that oxidative stress is a significant contributor in the pathogenesis of PE. Oxidative stress is defined as an imbalance between the generation of reactive oxygen species(ROS)and the capacity of anti-oxidants to prevent oxidative damage.Although PeroxiredoxinⅡis an important endogenous antioxidant,little has been known about it.Objective:To investigate the expression of peroxiredoxinⅡ(PrxⅡ)in placenta of PE and study its role in the course of PE.Methods:The expression of PrxⅡin placenta tissue was determined by western blot and compared between 15 cases severe pre-eclampsia patients and 15 cases normal pregnancies.The homogenates of placenta were extracted and the activity of superoxide dismutase(SOD)and the quantity of malondialdehyde(MDA),hydrogen peroxide(H2O2)were measured.Results:The expression of PrxⅡin severe pre-eclampsia was significantly lower than that in normal pregnancies(P<0.05).The activity of SOD of homogenate of placenta in severe pre-eclampsia was (89.74±18.26)U/mgprot,lower than control(105.23±21.88)U/mgprot (P<0.05)while MDA was(96.69±32.88)nmol/mgprot,higher than control.So as H2O2 in severe pre-eclampsia with the quantity (81.65±17.93)nmol/mgprot,which was higher than control(68.58±13.55) nmol/mgprot(P<0.05).Negative correlation between H2O2 and PrxⅡwas found both in study and control group.Conclusions:The expression of PrxⅡin pre-eclampsia decreased significantly and had a negative correlation between H2O2,which indicated that PrxⅡhas a close relation with the development of PE.
