Experimental Study On The Effect Of Shenditangmaining On The Immunity And Microvascular Complications In Diabetic Rats

Objective To find out the effect of Chinese traditional medicine Shenditangmaining on alteration of the immunity and occurrance of microvascular complications in diabetic rats.Methods The diabetic models in male SD rats are induced by intraperitoneal injection of Streptozotocin(STZ,60mg/kg)and are divided into: diabetes mellitus control group(oral dosing with same volume distilled water,n=12),aspirin group(oral dosing with aspirin,30mg/kg.d,n=12)and shenditangmaining group(oral dosing with medicinal broth of shenditangmaining containing 5g/kg.d,n=12).Take together with normal control group(n=12)that SD rats having the same age and body weight.To test weight and glycemia from vena caudalis blood weekly.the vena angularis blood are used to test CD3⁺,CD4⁺,CD8⁺T cells and CD4⁺/CD8⁺ ratio by Flow cytometry at the time point (before intervening and the end in 12_thweek);the same blood is centrifuged to detect serumTNF-αand IL-6 by ELISA at the same time piont above.At the time points of the beginning,the 6_thweek and the 12_thweek,the vena angularis blood is obtained and centrifuged to detect the serum triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),very low density lipoprotein cholesterol(VLDL-C).Thelast thymuses,pancreases,all the eyeballs and kindeys are selected to fix by 10%Formaldehyde and then to make them staining by HE,thymuses are weighing to test thymus index.Results The body weight is lower in DM group than in normal control group(P＜0.01),the body weight is more heavy in aspirin group and shenditangmaining group than in DM group(P＜0.05),but more light than in normal group(P＜0.01).In diabetes mellitus group,aspirin group and shenditangmaining group,the glycemia is all higher than in normal group(P＜0.01);but the glycemia is lower in aspirin group and shenditangmaining group than in diabetes mellitus group;the 9_thweek and 12_thweek,the glycemia in shenditangmaining group is lower than in aspirin group (P＜0.05).at the end of the study,the HbA_1cin diabetes mellitus group,aspirin group and shenditangmaining group are higher than in normal control group(P＜0.01),although HbA_1c in aspirin group and shenditangmaining group are not lower than in diabetes mellitus group (P＞0.05).The blood CD3⁺,CD4⁺,CD8⁺T lymphocyte are lower in diabetes mellitus group than in normal control group(P＜0.05);in aspirin group,CD3⁺,CD4⁺,CD8⁺T lymphocyte are higher than in diabetes mellitus group but CD3⁺,CD8⁺T lymphocyte are lower than normal control(P＜0.05 and P＜0.01);in shenditangmaining group,CD3⁺,CD4⁺,CD8⁺T lymphocyte is higher than in diabetes mellitus group(P＜0.05);but CD3⁺andCD4⁺ are not different from the normal control group(P＞0.05),CD3⁺,CD4⁺ in shenditangmaining group are higher than in aspirin group(P＜0.05).The 6_thweek and 12_thweek,serum TG,TC,LDL-C and VLDL-C in diabetes mellitus group are higher than in normal control group (P＜0.05);in aspirin group,serum TC,LDL-C and VLDL-C is lower than in diabetes mellitus group(P＜0.05)but serum TG is higher than in normal control group(P＜0.05);in shenditangmaining group,serum TG,TC,LDL-C and VLDL-C are lower than in diabetes mellitus group(P＜0.01);but not different from the normal control group(P＞0.05),the serum TG in shenditangmaining group is lower than in aspirin group(P＜0.05).Before starting,the serum TNF-a and IL-6 are higher in diabetes mellitus group,aspirin group and shenditangmaining group than in normal control group(P＜0.01);in the aspirin group,the serum TNF-a and IL-6 are lower than in diabetes mellitus group(P＜0.01);in shenditangmaining group,the serum TNF-a and IL-6 are lower than in diabetes mellitus group(P＜0.01)and are lower than in aspirin group(P＜0.05)but not different from the normal control group(P＞0.05),Compare to the normal control group,the thymus index decreased(P＜0.01);the thymus indexes in aspirin group and shenditangmaining group are increased(P＜0.01)and are not different from the normal control group(P＞0.05);With the light microscope,the construction of morphology of pancreases,thymuses,retina and kidneys in diabetes mellitus rats is derangement,inferior to the shenditangmaining group and aspirin group.The derangement of the retina improves significantly in shenditangmaining group.Conclusions Shenditangmaining improves the glycometabolism and lipid metabolism in diabetic rats,protects CD3⁺,CD4⁺,CDS⁺T cell and the thymus from impairing.Meanwile,it makes the serum TNF-a and IL-6 lower,slows the happen of the microvascular complications(retinopathy,cataract and diabetic nephropathy ect)in the diabetic rats.The medication of Shenditangmaining is more well than Aspirin.
