ObjectiveType 1 diabetic mellitus is a kind of autoimmunity disease whose feature is selectiveβcell destruction and high levels of blood glucose. In present, the main therapy is the usage of insulin and other drugs decreasing blood glucose. Many cytokines contribute to the pathogenesis and progression of type 1 diabetes mellitus (DM). Aspirin, cyclooxygenase inhibitor and anti-inflammatory medcine, used to prevent cardiovascular and cerebrovascular complications of DM. We used streptozotocin (STZ) to prepare the type 1 diabetic modle of rats. Then we used aspirin to cure the rats. Through analyzing the levels of plasma endothelin-1 (ET-1),pathological changes of hepatic tissue as well as Tumor Necrosis Factor-alpha (TNF-α) and interleukin-6 (IL-6) expression in liver of rats , we explored whether aspirin could be showed the protective action of vascular endothelial cell and liver damage with DM.Methods30 Sprague-Dawley(SD)rats were randomized into 3 groups: normal control rats(group NC), diabetic rats (group DM) and diabetic rats treated with aspirin(group AS). After 5 weeks of treatment, Blood glucose (BG), plasma insulin (INS) and ET-1 were measured, the pathological changes in the liver were observed under light microscope with hematoxylin and eosin (HE) staining, while TNF-αand IL-6 in liver were observed by immunohistochemistry.Results1. BG and ET-1 were higher in the diabetic rats than those in the normal control group (P<0.05), while INS was lower in the group DM ( P<0.05). Compared with the group NC , BG was higher (P<0.05), INS was lower(P<0.05) and ET-1 was the same (P>0.05) in the group AS.. There were no differences in BG, INS and ET-1 between group DM and group AS(P>0.05).2. Inflammatory lesions on the liver tissue were observed in group DM, while less changes in group AS.3. The expression of TNF-αand IL-6 in liver tissue were higher in the group DM than that in the group NC(P<0.05).Compared with group DM, the expression of TNF-αwas lower in group AS(P<0.05), while that of IL-6 was the same(P>0.05)。Conclusions1. Aspirin may not decrease blood glucose.2. Aspirin may not increase plasma insulin.3. Aspirin could play a protective role in vascular endothelial function in diabetic rats.4. Early and sustained intervention with aspirin may be useful to control inflammatory lesions on the liver tissue in diabetic rats.5. TNF-αand IL-6 play a role in the hepatic inflammatory lesions in diabetic rats. Aspirin can decrease the expression of TNF-α..
