The purpose of this paper was to study the preventive and therapeutic effects of Gushudan on osteoporosis in rats after administrated prednisolone and investigate the mechanisms initially.The results indicate that:Intramuscular inject prednisolone can increase the loss of bone mass,enhance bone absorption.Gushudan(3g/kg)can increase the body weight evidently after administrated for four weeks inrats;Gushudan(1~3g/kg)can conspicuous increase the bone mineral density and bending strength tested using dual X-ray bone density absorptiometry and 78X-2 type pellet four-use radiometer and also elevate the bone's wet weight and ash weight.Gushudan(0.3~3g/kg)can increase the contents of bone calcium; Gushudan(3g/kg)increase the contents of bone phosphorus evidently.Gushudan(3g/kg) distinguished decrease ALP contents and increase BGP contents in the serum.Gushudan(1~3g/kg)conspicuous depress the calcium contents in urine;At the same time,Gushudan (3g/kg)evidently increase cAMP and decrease cGMP in plasma.The results of microcirculation manifest that Gushudan(0.3~3g/kg)can enlarge the caliber,speed up flow rate and improve the state of microcirculation.Gushudan can significantly increase the proliferation of cultured UMR106.Therefore,Gushudan has a distinct preventive and therapeutic effect on osteoporosis rats caused by administration of prednisolone.The mechanisms maybe involve increasing bone mass,inhibiting bone resorption and increasing the proliferation of cultured UMR106.
