Constituents absorbed into blood of Divaricata Saposhnikovia Root (DSR) and its dynamic priciples are studied through Pharmacokinetics of TCM for setting a basis of the effective constituents.1.HPLC-fingerprint chromatograms of rat serum is established, fourteen constituents absorbed into blood after oral administration of DSR were detected, by comparing with the correspounding reference compounds, peak.5, 6, 8, 13 were preliminarily identified as prim-o-glucosylcimifugin, cimifugin, 5-o-methylvisamminol and sec-o-glucosylhamaudol. Relative retention time of all common peaks and relative peak area are calculated by the reference of cimifugin (peak.6). RSD of relative retention time of all common peaks is within 1%. Semblance is better than 0.99.2.10 constituents absorbed into blood are detected in rat plasma of 0-24 hours after oral administration of DSR, the principles of peak-area/time of such 10 compounds were elucidated. As a result, the principles of the 10 constituents are classified into three types by changing pathway. The peaks.2, 5, 7 and 8 will all showed a maximum absorption, and the properties of slowing absorption and rapid elimination were demonstrated. The pathways of peaks.3, 6, 11 showed two maximum absorption, the second maximum absorption is bigger than the the first, which shows that there may be other constituents transform into them. The plasma concentration reach the peak twice after the peak 10 absorbed into blood, which are induced by the hepatoenteral circulation of some active components. Part of cimifugin in the blood is from the original cimifugin absorbed into blood and part of cimifugin is from the transformation of prim-o-glucosylcimifugin and 5-o-methylvisamminol after metabolized in blood by analyzing the resource of cimifugin in blood.
