Objective: At the end of the last century, people found the Skp2 (S phase kinase-associated protein 2)being a new member of the proteinum family of F-box. According to the coding protein of p27kip1, however, p27kip1 , the dependence enzyme inhibitor of the cells through preventing cyclin and CDK forming compounds, interrupts the inversion of the cells from G1 stage to S stage. Based on the research,the high expression of Skp2, the low expression or the absence of p27kip1 are the significant reasons that the normal cells transform malignantly, propagate anomaly and differentiated, then are related with the formation, the progress of the malignant tumor and the degree of histological differentiation. Otherwise, according to the carcinoma of prostate, multiple myeloma, glioblastoma and cancer of larynx, the low expression or absence of PTEN (phosphatase and tensin homology deleted on chromosome ten)lowers with the expression of p27kip1 and increases with the expression of Skp2. So the anomal expression and the cooperation of three genes play an importantrole in the formation in filtration and aversion of the malignant tumor, and are related to the malignant differentiation of tumor cell. The paper adopts immunohistochemistry staining to detect the expression of Skp2, p27kip and PTEN in normal Intestine tissues, borderline tissues and Intestine Carcinoma tissues. It makes a preliminary research for the correlation of the three genes and study the relations between the three genes and clinicopathological parameters, and also makes a approach for the mechanism on carcinogenesis and development of the intestine carcinoma, accordingly, it dicusses the clinical significance in diagnosis, theraphy and prognosis of the intestine carcinoma.Meathod: The 54 fresh samples of the carcinoma of large intestine tissues coming from 54 the carcinoma of large intestine Patients who did not undertake chems and radiotherapy before the operation, have intact pathological datas, and the pathological type is glandular cancer. 12 samples are the borderline tissues, and 15 samples are normal intestine tissues, were soaked for about twenty-four hours by 10%formalin liquor, then paraffin imbedded to make blades, then they were stained with monoclonal antibodies. Through immunohistochemistry two-step method, detect the expression of the three proteins (Skp2,p27kip,pten)and the correlation among them were studied using statistic methods such as the chi-square test and Spearman correlation of ranks test,etc. P value less than 0.05 is considered to have statistic significance,and P value less than 0.01 is considered to be significantly different.Result:The expression of Skp2 in normal intestine tissues, borderline tissues,carcinoma of large intestine tissues and the relation with its clinicopathological parameters.The postive rates of Skp2 in normal mucosa tissues, borderline tissues,carcinoma of large intestine tissues are 0%, 33.33% and 48.15% respectively. The statistical results reveal that its postive rates in normal intestine tissues is significanly inferior to the one in carcinoma of large intestine tissues (p<0.01) and borderline tissues (p<0.05). However, the expression of Skp2 in carcinoma and borderline has no obvious difference. (p>0.05).The expression of Skp2 proteins in carcinoma of large intestine has no relations with the patient's age, sex, size (p>0.05), and has positive correlation with tumor differentiation (r=0.555,p<0.01), invasion of adventitia (r=0.463,p<0.01), lymph metabasis (r=0.295,p<0.05), distant metabasis (r=0.400,p<0.05).The expression of p27kip in normal intestine tissues, borderline tissues,carcinoma of large intestine tissues and the relation with its clinicopathological parameters.The postive rates of p27kip1 in normal mucosa tissues, borderline tissues, carcinoma of large intestine tissues are 100%, 83.33% and 51.85% respectively. The statistical results reveal that its postive rates in carcinoma of large intestine tissues is significanly inferior to the one in normal mucosa intestine tissues (p<0.01) and borderline tissues (p<0.05). However, the positive rate of p27kip1 in normal mucosa and borderline has no statistical signifance. (p>0.05).The expression of p27kip1 proteins in carcinoma of large intestine has no relation with the patient's age, sex, size, the invasion of adventitia and the distant metabasis (p>0.05),and has positive correlation with the tumor differentiation (r=-0.481, p<0.01) and lymph metabasis (r=-0.369, p<0.05) .The expression of PTEN in normal intestine tissues, borderline tissues,carcinoma of large intestine tissues and the relation with its clinicopathological parameters.The postive rates of PTEN in normal mucosa tissues, borderline tissues,carcinoma of large intestine tissues are 93.33%, 75% and 62.96% respectively. The statistical results reveal that its postive rates in carcinoma of large intestine tissues is inferior to the one in normal mucosa intestine tissues (p<0.05). However, the positive rate of PTEN in borderline and normal mucosa and the carcinoma of large intestine has no statistical signifance (p>0.05).The expression of PTEN proteins in carcinoma of large intestine has no relation with the patient's age, sex, size (p>0.05), and has negative correlation with tumor differentiation (r=-0.591, p<0.01), the invasion of the adventitia (r=-0.463, p<0.01), lymph metabasis (r=-0.388, p<0.05), distant metabasis (r=0.389, p<0.05).The correlation of the Skp2, p27kip1, PTEN proteins expression in carcinoma of large intestine tissues.The statistical results with the Spearman correlation of ranks test reveal that there is significanly negative corrleation between Skp2 and p27kip1 (r=-0.389, p<0.01), there is significanly negative correlation between Skp2 and PTEN (r=-0.410, p<0.01), and there is significanly positive correlation between p27kip and PTEN(r=0.575, p<0.01).Conclusion:The expression of Skp2 proteins in the carcinoma of large intestine may be related with tumor's proceeding and growth. Its high expression is the important definitive factor to the maligant transformation of the large intestine normal cells and metabasis of cancer cells in carcinoma of large intestine.The expression of p27kip1 proteins in the carcinoma of large intestine may influence tumor's proceeding and growth. Its low or absence expression may promote the maligant transformation of the large intestine normal cells and relate with the partial metabasis.The expression of PTEN proteins in the carcinoma of large intestine may influence tumor's proceeding and growth. Its low or absence expression may promote the maligant transformation of the large intestine normal cells and enhance cancer cells'infestation.With the depress of the differentiation degree of carcinoma of large intestine tissue, the high expresion of skp2 and the low expresion absence of the p27kip1 , PTEN are gradually obvious. So the anomal expressions of three genes influence the malignant differentiation of carcinoma of large intestine cells and are related with the malignant degree of the tumor.The dependability research of the expressions of skp2, p27kip1, PTEN in carcinoma of large intestine reveals that the process of carcinogenesis and the development of large intestine is the process that the oncogenes and anti-oncogenes interact and affect mutually. So the anomal expression and the dependability research of skp2, p27kip1, PTEN in the process of carcinogenesis, partial and distant transfer of carcinoma of large intestine reveal that the cooperation of three genes may affect the malignant transformation of the normal cells, then promote directly and indirectly the abnomal generation, differentiation of carcinoma of large intestine, and enhance its ability of partial and distant transfer and that it is significant to study the carcinogenesis, development and transfer of carcinoma of large intestine and appreciating its malignant degree and the prognosis.
