| Objective : To research whether there are differences between the the expression of PTEN in various pathological grading, and to investigate whether there are differences between the expressions of superficial bladder cancer and invasive bladder cancer. Also, whether there is any relationship between PTEN and Skp2,PTEN and Ki67 in the same organization. In addition, to research will the positive expression rate of PTEN of relapsed case be lower than that of recurrence free case. whether PTEN is related to patients'survival time after the operation.Methods: Choosing 10 cases of normal bladder which is confirmed by microscope with over 2.5cm far away from the tumors after total cystectomy; 47cases of transitional cell carcinoma samples are selected; PTEN, Skp2 and Ki67 were analyzed by way of Elivision-plus immunohistochemical technique. Four slices are trepanned from each specimen of paraffin blocks. Three of them are used to analyze PTEN,Skp2,Ki67, and another one is used to confirm the clinical stages and pathological grades of WHO with HE chromosome. Moreover, having following visited the patients'prognosis for 6 to 48 months. And then, to count the positive expressions of relapsed cases and recurrence free patients, to analyze Survival time after operation.Results:1,The positive expression rates of the normal bladder mucosa with PTEN and the pathological G1,G2, G3 are100% 85.6%﹑ 78.9%﹑ 46.7%, there are obvious differences between normal mucosa and G1(P<0.05), G1 and G3 , G2 and G3 lie significant differences as well(P<0.01), (P<0.05), respectively. On the contrary, there are no clear differences between G1 and G2.2,The positive rates of The PTEN in superficial bladder cancer and invasive bladder cancer have differences (P<0.05)。3,In the same bladder tissue, the expression of PTEN and Skp2 shows the negative correlation. (correlation coefficient r=-0.519 P<0.01)。4,In the same bladder tissue, the expression of PTEN and Ki67 shows the negative correlation. (correlation coefficient r=-0.657 p<0.01)。5,The positive expressions rate of the patients who relapse is much lower than those who are recurrence frees.6,Patients who show negative PTEN live shorter than those who show positive PTEN after operation.Conclusions The relationship between the biological characters of BTCC and the expression of PTEN and Skp2 is pretty close. The positive expression of PTEN and the grade malignancy of the BTCC reveal it negative correlation. PTEN and Skp2 may participate the biological characters of carcinoma of urinary bladder together. The positive expressions of the patients who do not relapse are obvious higher than those who relapse, which could be a standard to estimate whether the patients relapse easily or not after operation. PTEN also could be a criterion to assess how long can the survivors live. |
