The Research On The Relationship Between COMT Gene Polymorphism And The Susceptibility Of Endometriosis And Adenomyosis In Han People Of Hunan Province

OBJECTIVES: To study the association between polymorphism of COMT gene in exon 4 codon 1947(G→A) and the susceptibility to endometriosis and adenomyosis.METHODS: From 2007.4 to 2008.3,in the second affiliated hosipital of Xiangya medical school of Central South university, we collected 61 endometriosis cases(endometriosis group) and 40 adenomyosis cases (adenomyosis group) diagnosised by pathological section after given laparotomy or laparoscopy. Meanwhile we collected 67 women accepted oviduct anastomosis or accepted laparoscopic surgery as control group,we didn't find ectopic lesion or hysterauxesis.Using PCR-RFLP assay, we analysised the polymorphism of 1947 codon of COMT gene,then countered the frequency of the genotype and the allele to analysis if there exist association between COMT gene polymorphsim and the risk of endometriosis or adenomyosis. x~2 analysis was used to carry out data analysis using SPSS11.5 statistical software.To ensure the same genetic background, all objects came from Han people in Hunan province of China.RESULTS: The distribution of the genotype of COMT gene G1947A polymorphism of the three groups are accorded with Hardy-Weinberg equilibrium. There are no significant statistical difference(P>0.05) in the genotype and allele frequency of COMT gene between endometroisis group and control group. Wheras, between adenomyosis group and control group,there exists statistical difference. Compared with wide type genotype (G/G), the susceptibility of adenomyosis with heterozygote(G/A)and homozygote(A/A) may be increased by 3.125 and 4.375 fold.The allele A can increase the susceptibility of adenomyosis by 2.117 fold.CONCLUSIONS: For Han people of Hunan province,the polymorphism of COMT gene in exon 4 codon 1947(G→A) may have no association with endometriosis, but it may increase the risk of adenomyosis.
